高效液相色谱荧光检测法测定竹红菌中竹红菌甲素的含量
HPLC-FLD determination of hypocrellin A in Hypocrella bambusae(B.et Br.)Sacc
【摘要】 目的:建立高效液相色谱测定竹红菌中竹红菌甲素含量的方法。方法:采用 Zorbax Extend-C18柱(4.6mm×150mm,5μm),以甲醇与水(83:17,体积比)为流动相,流速1mL·min-1。荧光检测器激发波长为410nm,发射波长610nm。结果:竹红菌甲素在25~250μg·mL-1浓度范围内呈良好的线性关系(r=0.9999),平均回收率(n=6)为100.3%。结论:本分析方法简便快速,灵敏准确,可用于竹红菌中竹红菌甲素含量的测定。
【Abstract】 Objective:To establish an HPLC method for the determination of hypocrellin A(HA)in Hypocrella bambusae(B.et Br.)Sacc samples.Method:A Zorbax Extend-C18column(4.6 mm×150 mm,5μm)and a mobile phase consisting of methanol-water(83:17 by volume)were employed.The HPLC conditions were:flow- ing rate 1 mL·min-1,injection aliquot 10μL,column temperature 30℃,fluorescence detection excitation wave- length 410 nm and emission wavelength 610 nm.Results:HA showed good linearity(r=0.9999)in the range of 25-250μg·mL-1.The average recovery(n=6)was 100.3 %.Conclusion:The HPLC-FLD method is sim- ple,sensitive,accurate and rapid for the determination of HA in Hypocrella bambusae(B.et Br.)Sacc.
