毛细管电泳法测定虎杖药材中白藜芦醇苷、大黄素和大黄素甲醚的含量

【摘要】 目的:建立毛细管电泳法测定虎杖药材中活性成分白藜芦醇苷、大黄素和大黄素甲醚的含量。方法:采用 Agilent3DCE高效毛细管电泳仪,未涂渍熔融石英毛细管(75μm×48.5cm,有效长度40.0cm),压力进样5.25Pa×3s,运行缓冲液为10mmol·L-1硼酸钠溶液-甲醇(1:1),检测波长为220nm,运行电压30kV,温度25℃,内标为厚朴酚。结果:白藜芦醇苷、大黄素和大黄素甲醚浓度分别在20.5~410.8μg·mL-1(r=0.9990),9.78~156.4μg·mL-1(r=0.9998),12.13~121.3μg·mL-1(r=0.9992)的范围内呈良好线性相关;平均回收率分别为98.9%,99.0%,96.9%,RSD 分别小于1.8%,2.1%,1.6%。结论:该方法简便快速、准确、重复性好,可用于虎杖中自藜芦醇苷、大黄素和大黄素甲醚的含量测定。

【Abstract】 Objective:To determine the content of resveratrol-3-O-β-D-glucoside,emodin and rheochrysi- din in Rhizoma Polygoni Cuspidati by capillary zone electrophoresis.Method:Untreated fused silica capillary (48.5cm×75μm,effective length is 40cm)was used.Pressure injection of 5.25Pa×3s was employed,and the temperature was 25℃.The detection wavelength was set at 220nm.The samples were analyzed with 10 mmol· L-1sodium borate-methanol(1:1)running buffer at 30kV voltage.The internal standard substance was magnol- ol.Results:The calibration curve of resveratrol-3-O-β-D-glucoside,emodin and rheochrysidin showed good linearity over the range of20. 5-410. 8μg·mL-1(r=0. 9990),9.78-156.4μg·mL-1(r=0. 9998),12.13 -121.μg·mL-1(r=0. 9992),respectively.The average recoveries of the method were 98.9%,99.0% and 96.9%with RSD less than1.8%,2. 1%and1.6%,respectively. Conclusion:The method is simple,rapid,accu- rate,reproducible and can be applied in the determination of resveratrol-3-O-β-D-glucoside,emodin and rheochrysidin in Rhizoma Polygoni Cuspidati
