视网膜静脉阻塞家兔血管内皮生长因子的表达

【摘要】 目的研究血管内皮生长因子(Vascular Endothehal Growth Factor,VEGF)在视网膜静脉阻塞(RetinalVein Occlusion.RVO)中的作用及在视网膜新生血管中的参与机制。方法经玻璃体眼内电凝法建立 RVO 家兔模型,利用荧光素眼底血管造影(Funds Fluorescein Angiography,FFA)观察视网膜及血管的情况,采用免疫组化观察对照组、模型组视网膜组织中 VEGF 蛋白的表达情况。结果正常家兔视网膜组织中 VEGF 免疫反应弱。模型组与正常对照组比较,阻塞近端术后2 wk、4 wk,P<0.05(P=0.000,0.024),差异有显著性,而术后1 wk,P>0.05(P=0.144),差异不明显。各组两两比较,2 wk 组与1 wk、4wk 组,P<0.001(P=0.000,0.000),差异有显著意义。结论 VEGF 蛋白表达在视网膜局部缺血时提高,在再通或侧支循环形成时降低,与 RVO 眼内组织缺血形成存在一定的时空对应关系。

【Abstract】 Purpose To investigate the effect of VEGF in RVO and the possible mechanism in the pathogenesis of retinal neovascularization.Methods RVO of rabbit model was established with intraocular electrocoagulation to retinal vein next to the disc.Retinal vessels were observed by FFA,and VEGF expression was compared between the RVO model group and the normal control group by the Envision two-step immunohistochemical techniques.Results In normal control group、 VEGF protein was expressed weakly in partial cells of the inner retina and the retinal vessels almost negative stain.Compared to the controls,the expression of VEGF in the 2 th week and 4th week were significantly statistical difference,P<0.05(P =0.000,0.024).but there was no diferene in 1th week,P>0.05(P=0.144).The statistic study results showed P<0. 001,which means there was a significant difference between 2 th week and 1th week,4th week(P=0.000,0.1300). Conclusion The expression of VEGF was upregulated by retinal ischemica and fallen by collateralvessel formation or recanlization.It has the temporally and spatially correlation with RVO and might play an important role in the pathogenesis.
