水产品肌肉组织中氯霉素残留量检测方法的对比分析

【摘要】 初步比较了酶联免疫法(ELISA)中2种氯霉素试剂盒(荷兰EURO-DIAGNOSTICA试剂盒和德国r-Biopharm试剂盒)检测水产品氯霉素残留量的效果,并对测定水产品氯霉素残留量的气相色谱法(GC-ECD)和液相色谱—质谱法(LC-MS/MS)的技术参数进行了比较分析。结果表明,荷兰EURO-DIAGNOSTICA试剂盒和德国r-Biopharm试剂盒的检测范围分别为0.025~2.0μg·L-1和0.05~4.05 μg·L-1,其板内相对标准偏差均小于10%,2种试剂盒检测25个鱼或虾样品的假阳性率分别为12%和20%。LC-MS/MS和GC-ECD的最低检测浓度分别为0.1 μg·kg-1和0.27μg·kg-1,当氯霉素添加量为2 μg·kg-1时其平均回收率分别为95.2%和70.4%;LC-MS/MS样品前处理较简单,GC-ECD较繁琐。

【Abstract】 The examination effect of two ELISA kits (Netherlandish Euro-Diagnostica kit & German r-Biopharm kit) which were used to detect the chloramphenicol (CAP) residue of aquatic products such as fish and shrimp had been primarily compared. Further more , the comparative analysis of GC-ECD and LC-MS/MS was carried out. The results indicated that the detection range of Netherlandish Euro-Diagnostica kit and German r-Biopharm kit were 0. 025~2. 0μg·L-1 and 0. 05~4. 05 μg·L-1 respectively. The RSD of inter-assay was lower than 10%. The false positive rate of the two kits were 20% and 12% respectively when they were used to detect 25 fish and shrimp samples. The detection limits were 0. 1 μg· kg-1 and 0. 27 μg·kg-1 respectively. The recovery rates of standard sample added to 2 μg · kg-1 were 95. 2% and 70. 4% respectively. The sample pre-processing of LC-MS/MS was simple while GC-ECD was trivial.
