PLA表位肽免疫微球诱导的CTL对肝癌细胞的杀伤作用

【摘要】 目的考察载肽聚乳酸微球(PLA)体外诱导特异性CTL杀伤活性。方法采用标准51Cr释放法检测CTL杀伤活性。结果免疫做球M1、M2在体外均能刺激人PBMC增殖,形成大量可见克隆;CTL杀伤分析结果显示,免疫微球M1、M2诱导的效应细胞对荷肽T2细胞(T2+P)、HepG-2和Alexander的杀伤率均达75%以上,二者的杀伤活性没有明显差异(P>0.05)。它们对表达hAFP的肝癌细胞HepG-2和Alexander的杀伤率均显著高于不表达hAFP的膀胱癌细胞BTT和未荷肽的T2细胞,差异明显(P<0.001)。结论免疫微球M1、M2均能在体外诱导产生特异性CTL,并对表达靶抗原的肿瘤细胞有较强杀伤作用。

【Abstract】 Objective To survey the cytotoxicity of the CTL induced in vitro by PLA microspheres containing CTL epitopes derived from hAFP.Methods The specific cytolysis for hepatocellular carcinoma cell was assessed by 51Cr-release method.Results Two kinds of microparticles(M1,M2)containing CTL epitopes derived from hAFP induced HLA-A2+ PBMC from healthy volunteer to produce cell proliferation and form abundant visible clones;The cytolytic rate for hAFP+ tumor cells was over 75%.Which were significantly higher than HAFP- cells(P<0.001).And there was no significant difference between cytotoxicity of effector cells induced respectively by MI and M2(P>0.05).Conclusion Two kinds of microparticles have potent capabihty to induce specific CTL in vitro and kill hAFP+ tumor cells effectively.
