甘草酸单铵配伍复方丹参注射液预防大鼠子宫角粘连及对粘连局部tPA、PAI活性影响的研究

【摘要】 目的观察甘草酸单铵盐(mono ammonium glycyrrhetate,MAG)配伍复方丹参注射液对大鼠子宫角粘连的预防作用及其对大鼠腹腔液和子宫角粘连局部组织中tPA-a、PAI-a的影响。方法将大鼠分6组,制作大鼠子宫角粘连模型。MAG组给予MAG 5m/kg,复方丹参组给予复方丹参注射液1ml/kg,MAG加复方丹参组给予MAG 5mg/kg及复方丹参注射液1ml/kg,地塞米松磷酸钠(Dex)组给予Dex 1mg/kg,模型组给予生理盐水10ml/kg,空白对照组未实行任何处理。参照Leach的方法观察粘连评分、取腹腔液和局部粘连组织测组织纤溶酶原激活剂活性(tPA-a)和纤溶酶原激活抑制剂活性(PAI-a)。结果与模型组比较,MAG、MAG加复方丹参、Dex组明显降低粘连评分及严重粘连百分率(P<0.01),单用复方丹参未降低粘连评分。与模型组比较,复方丹参组与MAG加复方丹参组明显升高腹腔液tPA-a和tPA/PAI。与空白组比较,模型组明显升高了宫角组织中PAI-a、明显降低组织tPA/PAI,其它各实验组均明显升高组织tPA-a和tPA/PAI(P<0.01),且MAG加复方丹参组、Dex组明显降低组织PAI-a。结论本研究进一步证明了术后粘连形成过程包括局部纤溶活性降低过程的理论;证明了MAG和MAG伍用复方丹参具有预防粘连和提高纤溶活性的作用,且联合应用优于MAG单用。

【Abstract】 Objective To evaluate the effects of monoammonium glycyrrhetate(MAG)combined with com- pound Salvia Mihiorrhizae injection(CSM)on the prevention of uterine horn adhesion and the activity of tissue plasminogen activator(tPA-a)and plasminogen activator inhibitor(PAI-a)in hydrops abdominis and tissues of u- terine horn.Methods Rat models of uterine horn adhesion were established.The rats were divided into 6 groups, 10 in each.In the MAG group,the rats were given 5mg/kg MAG intraperitoneally;in the CSM group,1ml/kg CSM were given;in the CSM plus MAG group,1ml/kg CSM and 5mg/kg MAG were given;in the dexametha- sone(Dex)group,1ml/kg Dex was given;and in the model group,10ml/kg normal saline was given;while in the blank control group,no operation and medication were given.After 7 days,the rats were sacrificed to deter- mine the adhesion score referred to Leach method,adhesion tissue and peritoneal fluid were collected to determine the activity of tPA and PAI-a.Results Compared with the model group,the adhesion score was obviously reduced in CSM,CSM plus MAG,and Dex groups(P<0.01),CSM group could not reduce the adhesion score,CSM and MAG plus CSM groups obviously increased tPA-a and tPA/PAI in the peritoneal fluid.Compared with the blank control group,the model group increased PAI-a and reduced TPA/PAI in the adhesion tissue;other experimental groups obviously increased tPA-a and tPA/PAI in tissue(P<0.01);MAG plus CSM and Dex groups reduced tis- sue PAI-a obviously.Conclusion The results further proved the theory that post-operative adhesion formation in- cludes the lowering process of local tissue fibrinolytic activity,that MAG and MAG plus CSM could prevent adhe- sion and up-regulate the tissue fibrinolytic activity.Combined use of CSM and MAG is superior to that of using MAG alone.
