【摘要】 目的体外实验明确大蒜油能否增强肝枯否细胞(kupffer cell,KC)的抗肿瘤活性。方法采用酶消化法提取小鼠肝脏枯否细胞,分别以含大蒜油浓度为1200、600、300、150、75、37．5μg/ml的培养液培养枯否细胞24h。换洗培养液后,按10∶1效靶比加入小鼠结肠癌细胞CT26,共同培养24h。用MTT法测定活细胞数量,以未加药的KC/CT26共同培养和KC、CT26单独培养为对照组。结果KC的杀癌率为10．49%,经一定浓度大蒜油激活后,KC的杀癌率明显增强,并且杀癌率随大蒜油浓度的升高而增强,大蒜油浓度分别为1200、600、300、150、75、37．5μg/ml的杀癌率分别为91．91%、75．92%、73．71%、42．38%、37．94%和18．07%；300μg/ml组与600μg/ml组比较、37．5μg/ml组与不用大蒜油激活组比较差异均无显著性。结论大蒜油能激活肝枯否细胞,从而增强其抗癌活性。300μg/ml的大蒜油浓度可能是一个恰当的选择。更多还原

【Abstract】 Objective To investigate whether garlic oil has the effect in enhancing antitumor activity of kupffer cells(KCs)in liver of mice in vitro.Methods KCs were separated by enzymic digestion methods,and cultured with garlic oil in culture fluid containing different concentrations of 1 200、600、300、150、75、37.5μg/ml for 24 hrs rsrespectively.After renewal of culture medium,the KCs were cultured for another 24 hrs,together with mice colon carcinoma cells,CT 26 at the effect or/target ratio of 10:1.The number of survival KCs were measured with MTT,and compared to the control groups,including group of KC/CT26 culture without medication group, single KCs culture group and single CT26 culture group.Results The kill tumor rate of KCs was 10.49%,which could be significantly increase after activated by garlic oil in a concentration dependent manner.The kill tumor rate was 91.91%、75.92%、73.71%、42.38%、37.94% and 18.07% when the garlic oil at the concentration of 1 200、600、300、150、75、37.5μg/ml,respectively.But the difference of effect between the 300μg/ml and 600μg/ml garlic oil groups,and between the 37.5μg/ml garlic oil group and none garlic oil treated group was insig- nificant.Conclusion Garlic oil has an effect on activating liver KCs to increase its antitumor activity,and 300μg/ml is the optimal concentration.更多还原
