参麦注射液对大鼠脑出血后迟发性神经细胞损伤的保护作用研究
Study on Protective Effect of Shenmai Injection on Delayed Nerve Cells Injury of Rats after Intracerebral Hemorrhage
【摘要】 目的观察参麦注射液对大鼠脑出血后迟发性神经细胞损伤的保护作用。方法采用Rosen- berg法复制大鼠脑出血模型,观察脑出血后大鼠海马CA1区神经细胞病理形态结构、神经细胞线粒体DNA缺失、c-myc基因、PDGF-A基因表达及其参麦注射液的防治作用。结果参麦注射液减少大鼠脑出血后海马CA1区锥体细胞凋亡数目,抑制血肿周围脑组织线粒体DNA片段缺失,参麦注射液对出血早期c-myc mRNA的表达无明显影响,但可使PDGF-A mRNA表达明显下调并时限延长。结论参麦注射液可能通过调节PDGF表达对脑出血后迟发性神经细胞损伤修复有保护作用。
【Abstract】 Objective To observe the protective effect of Shenmai Injection(SMI)on delayed nerve ceils in- jury of rats after intracerebral hemorrhage.Methods Rosenberg method was adopted to duplicate rats’model of intracerebral hemorrhage.The pathological morphological structure,mitochondria-DNA(mtDNA)deletion,c-myc gene,platelet derived growth factor A(PDGF-A)gene expression in hippocampal CA1 area,and the protective effect of SMI were all observed.Results SMI could reduce the apoptosis number of pyramidal ceils in posthemor- rhagic CA,area,inhibit cerebral mtDNA deletion around hematoma.SMI showed no obvious effect on c-myc mR- NA expression in early hemorrhage,but it could obviously down-regulate PDGF-A mRNA expression and prolong the time of expression.Conclusion SMI has protective effect on delayed nerve cells injury after cerebral hemor- rhage by regulating PDGF expression.
