空气消毒剂过氧乙酸含量的酶催化光度法测定

【摘要】 过氧乙酸(PAcA)室内作消毒剂时,在空气中的含量很低,用通常的化学分析方法很难检测到.通过用过氧乙酸以喷雾法和熏蒸法进行空气消毒,用气体吸收管现场采样,以酶催化分光光度法测定室内环境中的微量过氧乙酸消毒剂的含量.实验证明:PAcA在辣根过氧化物酶(HRP)催化作用下,当磷酸盐缓冲液存在时能和苯酚、4-氨基安替比林发生Trinder反应.与过氧化氢的酶催化分光光度法相似,35℃生成的醌式染料30min显色完全,在505nm处有最大吸收,对应该波长的吸光系数为12 72L/(mmol·cm),测量过氧化物的工作曲线的线性回归方程为A=0 0038+12 72c(μmol·L-1),线性相关系数ρ>0 99,溶液组分(PAcA+H2O2)的检出浓度为3 09μmol·L-1,对应25mL过氧化物检出限为7 73nmol,过氧乙酸的检出限为0 235μg,发展了一种过氧乙酸痕量分析的简便、准确和灵敏的酶促反应显色方法.

【Abstract】 Peracetic acid, which is a kind of excellent disinfector, can be produced easily and kill many kinds of bacteria and virus effectively, SARS included.The indoor concentration of air disinfector is necessary to be detected online because of the erosion and toxicity.When peracetic acid is used as air disinfector of indoor, the concentration is too low to be measured by common methods of chemical analysis.The method of sprayer and fumigating method can be used when peracetic acid is used as air disinfector.Different concentrations of micro dosage of indoor peracetic acid absorbed by gas absorption tube are measured by biocatalytic spectrophotometry.Trinder coloring reaction of peracetic acid, phenol and 4-AAP, which is similar to that of hydrogen peroxide affecting horseradish peroxidase in existing phosphate buffer solution, are demonstrated in experiment and created the quinone dye matter. The biocatalytic photometry of color reaction at 35℃ in 30 min had the absorption peak at 505 nm, the absorptivity of corresponding wavelength of it is 12.72 L·mmol-1·cm-1.Equation of linear regression with workplot of determination of microscale peroxide is A=0.003 8+12.72 c(μmol·L-1) and linear regression coefficient ρ>0.99.The limit of detection is 3.09 μmol·L-1,to 25 mL solution component, corresponding limits of detections to peroxide disinfector and to peracetic acid are 7.73 n mol and 0.235μg.The selectivity of analysis is improved to determine the trace amount of peroxyacetic acid.The chromogenic reaction method catalyzed by HRP is employed to measure peracetic acid in the air by means of enzymatic photometry,showing its simplicity,accuracy and high sensitivity.
