Y染色体ZFY基因检测在组织工程中的应用

【摘要】 目的探讨人的Y染色体ZFY基因诊断在组织工程中的应用。方法对两种人的组织工程皮肤“替代物”进行RT-PCR检测,从mRNA水平检测Y染色体ZFY基因表达。用三条引物同时扩增,Y染色体可产生两个片断,X染色体产生一个片断。结果取男性细胞构建的表皮膜片修复女性皮肤缺损区的少量组织,可以检测到494bp和294bp两个条带;女性则494bp一个条带。检出率达100%(16/16)。用男性包皮角质干细胞构建的复合皮肤修复雌鼠全层皮肤缺损,取植入处少量组织检测到人的Y染色体494bp和294bp两个条带,且维持创面修复时间长达8个月。结论Y染色体ZFY基因诊断可作为组织工程皮肤替代物性别标记,且可以跟踪检测,具有特异、准确之优点。

【Abstract】 Objective To study the diagnosis of ZFY gene on human Y chromosome and its application in tissue engineering. Methods To detect two tissue-engineered skin substitutes with RT-PCR (Reverse transcript-polymerase Chine reaction). It can be found that ZFY gene of human Y chromosome expression at mRNA level. Amplify with three primers at same time. Two bands (494bp; 294bp) would be detected from Y chromosome; one band (494bp) form X chromosome. Results It showed that two bands (494bp; 924bp) occurred at the sample of male tissue engineered skin membrane after transplantation; one band (494bp) occurred from female. The rate of detecting is 100% (16/16). The bilayered tissue-engineered skin with keratinocyte stem cell of male children could be used to repair the skin defect of female rat. The results of RT-PCR suggested that Y chromosomes could be detected in the samples after three months and six months still visible to eight months. Conclusion Diagnosing of ZFY gene on human Y chromosome as a sex marker in tissue engineering is very specific and accuracy.
