螺旋藻中β-胡萝卜素高效液相色谱测定方法的研究
Study on HPLC Method for Detecting β-Carotene In Spirulina platensis
【摘要】 采用高效液相色谱法直接测定螺旋藻中β-胡萝卜素组分的含量。色谱柱为YMC-pack ODS(4.6mmi·d×150mm,5μm),流动相为甲醇-乙腈-二氯甲烷(体积比60∶30∶10),在435nm波长处检测。研究结果表明:β-胡萝卜素的检测限为20ng,线性范围在4.60~36.80mg/L,相关系数为0.9996,加样平均回收率为96.61%。方法准确、简便、快速,适用于螺旋藻及螺旋藻制品中β-胡萝卜素组分的检测。
【Abstract】 The high performance liquid chromatographic (HPLC)method for determination of β-carotene in Spirulina platensis was developed.The chromatographic conditions were as follows:YMC-pack ODS ( 4.6 mm i.d× 150 mm column, 5 μm ),V(methanol)∶V(acetonitrile)∶V(H2O)=60∶30∶10 mobile phase and UV detector at 435 nm .The β-carotene in myocardium was extracted with 20 ng .The calibration curve showed a good linearity (r= 0.999 6 ).And the average recovery of the method was 96.61%.The rapid and accurate method is particularly suitable for the determination of β-carotene in spirulina.
