磷酸氟达拉滨的HPLC测定
Determination of Fludarabine Phosphate by HPLC
【摘要】 目的:建立用HPLC测定磷酸氟达拉滨的方法。方法:采用YMGODS-AQ色谱柱,梯度洗脱法,A泵用0.68%的KH2PO4的水溶液(用磷酸调pH至3.0),B泵为50%(V/V)乙腈水溶液,流速为1.0ml·min-1,检测波长为263nm。结果:磷酸氟达拉滨在10-80μg·ml-1范围内线性关系良好(r=0.9998),平均回收率为99.6%,RSD=0.5%(n=5)。结论:该方法准确、可靠,灵敏度高,重复性好。
【Abstract】 Objective: A HPLC method for determination of fludarabine phosphate was established. Method: A YMG ODS-AQ column was used, the mobile phase was pump A-0. 68% monobasic potassium phosphate (pH adjusted to 3.00 with phosphoric acid) and pump B-50% (V/V) acetonitrile water solution, the flow rate was 1.0 ml ·min -1 , using gradient elution method the detecting wavelength was at 263 nm. Result: The calibration curve was linear within a range of 10-80 μg ·ml-1 ( r = 0. 999 8 ) . The average recovery was 99.8% with RSD of 0.8% (n =5). Conclusion: The method is accurate, reliable, sensitive, and repeatability.
