硫糖铝壳聚糖凝胶的制备及小鼠的急性毒性试验

【摘要】 目的:探讨硫糖铝壳聚糖凝胶的制备方法及对小鼠的急性毒性。方法:通过正交实验对硫糖铝壳聚糖凝胶处方进行筛选;用硫糖铝壳聚糖凝胶对小鼠灌胃给药,进行小鼠的急性毒性试验。结果:通过实验筛选,拟定出硫糖铝壳聚糖凝胶的处方为每1000ml含硫糖铝100g;壳聚糖3g;甲基纤维素4g;糖精钠0.2g;聚山梨酯8020g;10%尼泊金甲酯和尼泊金乙酯乙醇溶液(1:1)8.33ml;水适量。结论:选定的处方及制备工艺是可行的,硫糖铝壳聚糖凝胶对小鼠的急性毒性较低。

【Abstract】 Objective: To study preparing method and acute toxicity to mice of sucralfate chitosan gel. Method: By orthogonal experiment, the ingredients of sucralfate sucralfate gel prescription were selected and its acute toxicity to mice were tested by feed. Result: The selected prescription of sucralfate gel were: sucralfate 100 g, water soluble chitosan 3 g, methylcellulose 4 g, methylparaben and ethylparaben(1:1)0. 833 g; saccharin sodium,0. 2 g; tween 80 20 g, water 900 ml. The ratio of sedimentation to volume for the prescription was 0.97 in 3 hours. It would be uniform if it was laid aside for 48 hour and reversed for 3-4 times. LD50 were not able to be obtained if administration ig because there was not any mouse died. The largest amount hy ig for mouse equaled to 200 times for human dose. Conclusion: The prescription that has been selected is fit for the demand of sucralfate chitosan gel and its acute toxicity are relatively low.
