芪棱汤对大鼠局灶性脑缺血再灌注后核因子-κB表达的影响
The Effects of Qileng Decoction(芪棱汤 )on Expression of Nuclear Factor-κB in Rats with Focal Cerebral Ischemia-Reperfusion
【摘要】 目的观察芪棱汤对大鼠脑缺血再灌注后核因子-κB(NF-κB)表达的影响。方法雄性SD大鼠随机分为正常组、假手术组、模型组及治疗组,除正常组外其余各组按再灌注的时间又随机分为1、3、6、12、24、48和72h共21个亚组;采用颈内动脉线栓与环扎法建立MCAO模型。在术前12、24h及术后即刻治疗组予腹腔注射芪棱汤精制液,余组给予等体积生理盐水。动物杀检前进行神经学评分;采用免疫组化二步法检测再灌注各时点NF-κBp65和ICAM-1的表达。结果正常组和假手术组神经学评分正常,治疗组神经学评分较模型组显著降低。NF-κBp65在正常组和假手术组有微弱表达,两组情况相近;模型组和治疗组NF-κBp65表达较假手组明显增加,但治疗组表达低于对照组。ICAM-1在正常组和假手术组未见表达,治疗组和模型组表达增加,与模型组相比,治疗组ICAM-1表达显著减少。结论芪棱汤可降低NT-κB和ICAM-1的表达,可能是其抗脑缺血再灌注损伤的部分作用机制。
【Abstract】 Objective:To investigate the effects of Qileng Decoction(芪棱汤 ) on expres sion of nuclear factor kappa B (NF- κ B)in rats with focal cerebral ischemia- reperfusion.Methods:Male SD rats were ramdomly divided into four groups;the nat ive group(n=6), the sham- operation group(n=42),the MCAO group(n=42) and the t reatment group(n=42),and the last three groups were further divided into seven s ub- groups respectively according to the reperfusion time of 1,3,6,12,24,48 and 72 hours(n=6 for each sub- group).Rat model of middle cerebral atery occlusion (MCAO) and recanalization was made with thread liation.24h 12h before and right after the operation,the treatment group were intraperitoneally injected with the refining solution of Qileng Decoction while other groups were injected with nor mal solution.Neurologic scoring was made before rats were sacrificed and NF- κ B P65 and ICAM- 1 were detected with immunohistochemistry.Results:There were n o changes in neurologic scoring in the native and sham- operation group which d ramatically increased in the MCAO group and treatment group.Scores in the treatm ent group significantly decreased compared in that in the MCAO group.Weak expre ssion of NF- κ B P65 was observed in the native and sham- operation between w hich there was no statistical difference, while in increased in MCAO and treatm ent group compared with that in sham- operation group and decreased in the trea tment group compared with that in MCAO group.There was no observed ICAM- 1 expr ession in the native and sham- operation group which increased in the MCAO grou p and treatment group and decreased in the treatment group with comparison to th at in the MCAO group.Conclusion:Qileng Decoction decreases the expression of NF - κ B and ICAM- 1 in rats after cerebral ischemiareperfusion,which partially explains the mechanisms of Qileng Decoction against cerebral ischemia- reperfus ion injury.
