【摘要】 目的观察葛根素对球囊损伤血管内皮后血小板活化的影响。方法84只雄性Wistar大鼠随机分为假手术组、手术组和葛根素组各28只,手术组与葛根素组建立大鼠球囊损伤主动脉内皮模型,葛根素组于术前开始每日腹腔注射葛根素(50mg/kg·d)。各组分别于术后第3、7、14日采血,采用抗人活化型血小板表面P-选择素特异性单克隆抗体,以放射免疫法测定各时间点的血小板活化状态;放射免疫法测定血浆TXB、26-keto-PGF1α的浓度。结果球囊损伤主动脉内皮后第3日手术组血小板表面P-选择素较假手术组明显升高,第7日开始下降,第14日恢复至术前水平;葛根素组较手术组各时间点均降低,与假手术组相近。手术组与假手术组比较,内皮剥脱后第3、7日时血浆TXB升高,第14日时差异不显著;葛根素组较手术组在各2时间点血浆TXB水平均降低,与假手术组相近。手术组血浆6-keto-PGF1α水平则较假手术组降低,在术2后第3、7日差异显著,至第14日水平相近;葛根素组较手术组血浆6-keto-PGF1α水平明显升高,以第3、7日明显,第14日时与手术组和假手术组均相近。结论球囊损伤血管内皮后血小板在至少7d内处于活化状态,葛根素能抑制血小板的活化,减少血小板与内皮下层的粘附,从而在一定程度上起到预防血栓形成、避免栓塞的作用。更多还原

【Abstract】 Objective:To investigate the effect of Puerarin on platelet activation after balloon angioplasty.Methods:84 wister rats were randomly devided into 3 groups,Group 1(n=28) served as the controls,Group 2(n=28) were given vascular balloon injury,Group 3(n=28) were given Puerarin A Study on the Effect of Puerarin on P-Selection,TXB2 and 6-keto-PGF1αActivation in Rat Model after Balloon Angioplasty$$$$ Ji Wenyan,et al Objective:To investigate the effect of Puerarin on platelet activation after balloon angioplasty.Methods:84 wister rats were randomly devided into 3 groups,Group 1(n=28) served as the controls,Group 2(n=28) were given vascular balloon injury,Group 3(n=28) were given Puerarin [50mg/(kg·d),ip.) from 1 day before injury to 14 days after injury.The levels of plasma TXB2 and 6-keto-PGF1αand the number of P-selection molecule on platelet surface were measured at the day 3,7 and 14 after balloon injury.Results:(1)The number of platelet membrane P-selection of Group 2 were obviously higher than those of Group 1 atfer balloon injury at the day 3.The number began to go down after 1 week and reached the same level as Group 1 till 2 weeks.The number of platelet membrane P-selection of Group 3 were lower than that of Group 2 at every time.There were no difference between Group 3 and Group 1.(2)The level of Group 2 of plasma TXB2 were higher than that of Group 1 at the day 3 and 7 after balloon injury.There were no difference between Group 2 and Group 1.The level of plasma TXB2 of Group 3 were lower than Group 2 at every time and had no difference with Group 1.(3)The level of plasma 6-keto-PGF1αof Group 2 were lower than that of Group 1.The difference were obviously at the day 3and 7 and there were no difference till the day 14.The level of plasma 6-keto-PGF1αof Group 3 were obviously higher than that of Group 2,especially at the day 3 and 7.There were no difference between Group 2 and Group 3 after 2 weeks.Conclusion:The platelets are at the state of activation at least one week after balloon injury.Puerarin can lower platelet activation and decrease the adhesion between platelets and intimal.Puerarin contribute to antithrombotic and avoiding thrombotion to a certain extent.更多还原
