【摘要】 目的:进行当归药材的质量标准研究,着重建立当归药材的TLC鉴别和阿魏酸的含量测定方法。方法:采用TLC法定性鉴别当归药材;采用HPLC法建立当归药材中阿魏酸的含量测定方法。色谱条件:①HPLC色谱条件:色谱柱:ZorbaxEclipseXDB-C18(4.6×150mm,5μm);流动相:甲醇-水-冰醋酸(30:70:0.2);流速1.0mL/min;检测波长:320nm;柱温25℃。②TLC条件:采用硅胶GF254板,以正己烷-氯仿-冰醋酸(6:4:1)为展开剂,CAMAGⅢ型薄层扫描仪紫外365nm数码摄像。结果:HPLC法阿魏酸在20ng～200ng范围内,线性关系良好,回归方程为Y=5649.0267X+0.0000(r=1.000);平均加样回收率为100.21%,RSD=1.80%(n=6),十批当归药材阿魏酸的含量在0.09～0.33mg/g。TLC鉴别结果满意。结论:建立了当归药材的TLC鉴别方法和阿魏酸的含量测定方法,并制定了阿魏酸的含量限度,方法操作简便、准确快速、重现性好,可完善现行2000年版中国药典,有效控制当归药材的质量。更多还原

【Abstract】 Objective:To establish the quality standard for Danggui,especially to develope a method for determination the content of feruLic acid in Danggui by HPLC and TLC identified method.Methods:The content of feruLic acid in Danggui were determined by HPLC.HPLC Condition:Zorbax Eclipse XDB-C_ 18(4.6×150mm,5μm);the mobile phase is methanol-water-HAc(30:70:0.2) at 25℃,the flow rate was 1.0 mL/min,and UV detection wavelength was at 320nm.Danggui were identified by TLC.The TLC Condition:GF254 as the stationary phase,the cyclohexane-chloroform-glacial acetic-acid(6:4:1) as the mobile phase,detected by UV(365nm).Results:The contents of ferulic acid were determined by HPLC There was a good linear relationship between the absorption peak area and the concentration at the rang of 20ng～200ng,Y=5649.0267X+0.0000(r=1.000)。The average recovery was 100.21%,with RSD was 1.80%.Ten Danggui samples of Danggui which bought from different areas were detected,the contents of feruLic acid in Danggui were from 0.09mg/g to 0.33mg/g.Danggui could be identified by TLC.The TLC sports developed was fairly clear.Conclusion:The method to identifiy Danggui by TLC were established.The method for determination of feruLic acid in Danggui by HPLC was established.The content limit of feruLic acid in Danggui was developed.The method is simple,accurate,rapid,reliable,and reproducible.And can be used to control the quality of Danggui effectively.更多还原
