mIL-12基因转染J558肿瘤细胞疫苗的抗瘤实验研究
Experimental Research on Antitumor Effect of J558 Tumor Cell Vaccine Transfected by mIL-12 Gene
【摘要】 目的 探讨mIL -12基因转染的肿瘤细胞疫苗的抗肿瘤作用。方法 将鼠源性mIL -12基因转染J5 5 8细胞制备工程瘤细胞后免疫 2 0只BALB/c小鼠 ,另选 2 0只小鼠为对照组 ,用ELISA法测定J5 5 8/mIL -12瘤细胞培养上清液及对照组中mIL -12水平。经工程瘤细胞免疫的小鼠淋巴结细胞与J5 5 8肿瘤细胞共同培养后用ELLSA法测定其上清液中mIFN -γ水平。用J5 5 8瘤细胞攻击免疫小鼠及对照组小鼠以观察其存活情况。结果 J5 5 8/mIL -12瘤细胞培养上清液中mIL -12水平显著高于对照组 (895 2± 2 8 4vs 6 64± 1 5 3pg/ml,P =0 0 0 0 ) ;免疫小鼠淋巴结细胞与肿瘤细胞共同培养后上清液mIFN -γ水平显著高于对照组 (997 61±116 3 6vs 6 3 6± 1 2 7pg/ml,P =0 0 0 0 ) ;CTL细胞毒实验证实免疫小鼠CTL杀伤J5 5 8肿瘤细胞为特异性杀伤 ;特异性肿瘤保护试验显示该型瘤苗具有良好的抗瘤作用。结论 mIL -12基因转染的J5 5 8肿瘤细胞疫苗具有良好的抗瘤作用。
【Abstract】 Objective To study the antitumor effect of J558 tumor cell vaccine transfected by mouse interleukin-12 (mIL-12) gene. Methods J558 tumor cells were transfected by mIL-12 gene to develop an engineered tumor vaccine, which were used to vaccinate 20 BALB/c mice. Another 20 BALB/c mice were chosen as blank control group. The mIL-12 levels in two groups were measured by enzyme-linked immunosorbent assay (ELISA) method. Lymphoid nodule cells from experimental group were co-cultured with J558 cells to detect the mouse interferon-γ (mIFN-γ) level in culture supernatants. The survival data were compared in two groups with J558 tumor cells assault. Results The mIL-12 level in experimental group was significantly higher than that in control group (895.2±28.4 vs. 6.64±1.53pg/ml, P=0.000). The mIFN-γ level was also significantly higher than that in control group (997.61±116.36 vs. 6.36±1.27pg/ml, P=0.000). CTL cytotoxicity experiment confirmed that it was specific for CTL against J558 cell in immunized mice. The specific protection experiment against tumor showed that J558 tumor cell vaccine transfected by mIL-12 gene had good antitumor effect. Conclusion J558 tumor cell vaccine transfected by mIL-12 gene induces efficient antitumor immunity.
