【摘要】 目的:探讨党参单味药提取物对大鼠壁细胞胞内游离钙离子浓度的作用。方法:取SD大鼠连续给党参提取物5天,每天一次,末次给药后两小时心脏采血制备含药血清;将分离后壁细胞用荧光指示剂-Flou3-AM负载,然后用共聚焦显微镜检测胞内游离钙离子浓度;钙离子浓度以荧光强度(FI)表示。结果:党参药物血清高、中剂量组与对照组相比均有显著性差异(P<0.05)。结论:党参药物血清可剂量依赖性地减少胃泌素刺激的壁细胞胞内游离钙离子浓度的增高,这可能是我们以往发现它减少胃酸分泌而治疗溃疡性疾病的机制之一。更多还原

【Abstract】 Objective: To study the effects of Dangshen root extract on intracellular free calcium concentration [Ca2 +]i of parietalcells.Methods: The Dangshen-containing serum was obtained from rat blood after a continuously five days’feeding with Dangshen rootextract of three doses and parietal cells were isolated from Sprague-Dawley rats;[Ca2 +]i in single cells was measured by confocal mi-croscope loaded with Fluo3-AM as fluorensent indicator;The change of [Ca2 +]i was represented by fluorensent intensity(FI).Re-sults: There were differences in the FI of [Ca2 +]i increased by gastrin both between high and control group and between middle andcontrol group(P <0.05).But no difference was found between lowand control groups(P >0.05).And time to peak of FI,it was notfound any difference between any two of the groups.Conclusion: Dangshen-containing serum can inhibit the intracellular [Ca2 +]i in-crease induced by gastrin in a dose-dependent manner and it may be one of the mechanisms of its reduction on acid secretion which hasa close relation with the formation of ulcerous diseases.更多还原
