益活清胰汤对重症急性胰腺炎大鼠海马CAI区嗜碱性成纤维细胞生长因子的影响

【摘要】 [目的]探讨益活清胰汤对大鼠重症急性胰腺炎(SAP)并发胰性脑病(PE)海马CAI区嗜碱性成纤维细胞生长因子(bFGF)的影响。[方法]45只SD大鼠建立SAP模型,随机分为益活清胰汤(C)组、造模(B)组、假手术(A)组,测定6、12、24h各时点血浆肿瘤坏死因子α(TNFα)、白细胞介素6(IL6)、磷脂酶A2(PLA)值。对大脑皮层和海马组织形态、bFGF的表达进行观察。[结果]B、C两组术后6hTNFα、IL6、PLA2开始升高,24h达高峰。C组6、12、24h各时点的TNFα、IL6、PLA2值小于A组(P<0.01)。海马bFGF表达显著增多。[结论]益活清胰汤能降低SAP血浆TNFα、IL6及PLA2水平,改善脑血流量,对抗缺血低氧,增强SAP时海马组织bFGF的合成、分泌,阻止神经细胞凋亡和(或)促进新的神经元出芽及突触形成。

【Abstract】 [Objective] To study the effect of Yihuo Qingyi decoction on basic fibroblast growth factor (bFGF) neuron in hippocampus CAI area of rats with severe acute pancreatitis (SAP) combined with pancreatic encephalopathy (PE).[Methods] Forty-five SD Rats were induced SAP and divided into Yihuo Qingyi decoction treatment group (group C), model group ( group B) and sham operation group (group A). Blood was obtained respectively 6 hrs, 12 hrs, 24 hrs after operation. Expression of TNF-α, IL-6 and PLA was measured. Pathomorphologic changes of cerebral cortex and hippocampus were observed. Positive basic fibroblast growth factor (bFGF) neurons in hippocampus were determined by immunohistochemical method and the expression of bFGFmRNA in hippocampus was measured by in situ hybridization. [Results] In the group B and C TNF-α, IL-6, PLA2 began to rise 6 hrs after operation, reached the peak 24 hrs after the operation. In the group A 6 hrs, 12 hrs, 24 hrs after operation, TNF-α,IL-6 and PLA2 were lower than those in the group B (P<0.01). Expression of bFGF in hippocampus increased obviously (P<0.01).[Conclusion]Yihuo Qingyi decoction can lower the serum levels of TNF-α, IL-6 and PLA2 in SAP, regulate brain blood infusion and resist ischemia and hypoxia, upgrade the synthesis and release of bFGF in hippocampus, inhibit neuron apoptosis and stimulate neurons reproduction.
