免疫检测用纳米胶体金的制备及粒径控制

【摘要】 利用柠檬酸钠作为还原剂,采用液相还原法制备了适用于免疫检测的胶体金纳米颗粒。用透射电子显微镜和紫外/可见光分光光度计分别对胶体金颗粒的形貌和光学吸收特性进行了检测。结果表明,柠檬酸钠的加入量是影响胶体金颗粒粒径和形貌的主要因素,制备100ml0.01%胶体金溶液,1%柠檬酸钠溶液加入量小于5ml时,胶体金颗粒的平均直径随柠檬酸钠加入量的增加而减小,当大于5ml时,胶体金颗粒的平均直径随柠檬酸钠量的增加而增大。在研究范围内,胶体金颗粒的平均粒径与最大吸收峰波长之间存在直线回归关系。制备的胶体金呈单分散的球形分布,具有较高的生物学活性,适用于胶体金免疫检测。

【Abstract】 Colloidal gold nanoparticles, which are used for immunity test, were prepared through liquid reduction with sodium citrate as reducing agent. Transmission electron microscopy (TEM) and UV/visible absorption spectrophotometry are used to check the shape and the optical absorption of colloidal gold. It shows that the amount of sodium citrate is the main influencing factor to the size and shape of the colloidal gold. The mean diameter of the colloidal gold increases with the decrease of the sodium citrate adding amount if the total amount of sodium citrate is less than 5 ml. The mean diameter of the colloidal gold increases with the increase of the sodium citrate adding amount if the total amount of sodium citrate is more than 5 ml. Within the range of our research, there exists the linear regression relationship between the mean diameter and the wavelength of maximum absorbance. The colloidal gold presents monodispersity and spheric shape and has good biological activity.
