异丙酚对睾丸扭转/复位后生精细胞凋亡及相关基因表达的影响鄢
Effects of propofol on apoptosis of rat testicular germ cells and expressions of Bcl-2 and Bax gene induced by testicular torsion/detorsion
【摘要】 目的探讨异丙酚对睾丸扭转/复位后睾丸细胞凋亡及相关基因表达的影响。方法30只青春期Wistar大鼠随机等分为3组。组1为对照组(左侧睾丸扭转720°即刻复位);组2、3为睾丸扭转/复位模型组(左侧睾丸扭转720°、2h后复位)。组3睾丸复位前5min静脉注射异丙酚20mg/kg,继之输入5%异丙酚50mg/(kg·h)1h;组1、2以生理盐水代之。术后24h取左侧睾丸标本分别以原位缺口末端标记法(TUNEL)及免疫组织化学法检测凋亡细胞和Bcl-2、Bax基因蛋白表达。结果与组1比较,组2睾丸生精细胞凋亡指数升高和Bax表达增强(P<0.01),而睾丸细胞Bcl-2表达下降(P<0.01),Bcl-2/Bax比值降低,睾丸病理损害明显。与组2比较,组3睾丸上述改变明显减轻(P<0.01)。结论异丙酚通过调节Bcl-2及Bax蛋白表达抑制睾丸扭转/复位所诱导的生精细胞凋亡增加。
【Abstract】 [Objective] To investigate the possible protective role of propofol on apoptotic cell death and expressions of Bcl-2 and Bax gene proteins in a model of torsion/detorsion of the rat testis.[Methods] Thirty adolescent male Wistar rats were randomized into three groups of ten rats each.Group1 the left testis underwent a clockwise 720° torsion then untwisted at once as a control;in group 2,3 the left testis underwent a clockwise 720° torsion for 2 h followed by detorsion;in group 3 propofol was dripped intravenously 5 min before the left testicular detorsion and then kept 1 h.24 h after operation,left testis was removed and assessed for cell apoptosis and expressions of Bcl-2 and Bax by TUNEL technique and immunohistochemical method respectively.[Result] In group 2,the germ cell apoptotic index and Bax expression of the testis were significantly increased compared with those in group1(P <0.01),while Bcl-2 expression decreased obviously(P <0.01),the ratio of bcl-2/Bax decreased and the pathological lesions of the testicle were obvious.The changes mentioned above were alleviated obviously in group 3 compared with those in group 2(P <0.01).[Conclusions] Propofol can suppress the apoptosis of germ cells induced by testicular torsion/detorsion through its regulation of Bcl-2 and Bax gene protein expressions,which may be important for protecting the testis from torsion/detorsion injury.
