CD8+细胞强化骨髓细胞输注免疫调节效应及机制

【摘要】 目的观察不同脾细胞亚群强化骨髓细胞输注调节特异性免疫状态的效应,并探讨其可能机制。方法C57BL/6小鼠分别输注BACB/C小鼠5×106骨髓细胞、5×106骨髓细胞+5×106脾细胞、5×106骨髓细胞+5×106CD4+细胞和5×106骨髓细胞+5×106CD8+细胞,3月后观察特异性免疫状态和淋巴细胞分化状况。结果CD8+细胞联合输注的受体淋巴细胞反应较弱,IL-2和IFN-γ低表达,IL-4高表达。结论CD8+细胞输注强化了骨髓细胞输注调节特异性免疫状态的效应,包括免疫偏移在内的机制参与了这一过程。

【Abstract】 [Objectives] To investigate the effect and the mechanisms of subtypes of splenocytes improving the regulatory effect on specific immune response following donor bone marrow infusion. [Methods] C57BL/6 mice were infused cells from BACB/C mice: 5×106 bone marrow cells, 5×106 bone marrow cells +5×106 splenocytes, 5×106 bone marrow cells +5×106 CD4+ lymphocytes or 5×106 bone marrow cells +5×106 CD8+lymphocytes.3 months later the donor specific immune response and the cytokine profile were observed. [Results] The CD8+ combined infusion but not the other treatments regulated the receipient’s specific immune response compared with the bone morrow single infusion, and the expression of IL-2 and IFN-γ decreased while the expression of IL-4 increased. [Conclusions] CD8+ lymphocyte infusion improves the regulatory effect on specific immune response following donor bone marrow infusion. Several mechanisms including immune deviation involve this process.
