P53和PCNA在辐射诱导犬胆管增殖平滑肌细胞凋亡中的表达及意义

【摘要】 目的探讨在辐射诱导犬胆管增殖平滑肌细胞凋亡中,P53和PCNA的表达及意义。方法12只犬随机分两组,每组6只犬,在犬胆管内分别植入103pd(103钯)放射性支架和普通胆管支架,术后30d取出胆管标本采用ABC法进行PCNA免疫组化染色,核出现棕色反应为阳性细胞;原位杂交进行P53细胞染色,细胞浆出现棕褐色反应为阳性细胞;又行平滑肌细胞凋亡染色,阳性细胞为细胞核有棕黄色颗粒即为凋亡细胞。结果103Pd支架组PCNA蛋白阳性表达率明显低于普通支架组,两组之间比较差异有非常显著性(P<0.01),103Pd支架组P53蛋白阳性表达率明显高于普通支架组,两组之间比较差异有非常显著性(P<0.01)。103Pd支架组平滑肌细胞凋亡数较普通支架组明显增多,两者有显著差异(P<0.05),103pd放射性支架组犬胆管腔无明显狭窄,而普通支架组犬胆管腔有明显狭窄。结论以上结果证实103Pd支架组犬胆管平滑肌组织PCNA表达较弱,而普通支架组犬胆管平滑肌组织PCNA表达却增高,说明辐射可以抑制平滑肌细胞增殖,辐射可激活P53基因而诱导犬胆管平滑肌细胞凋亡,同时抑制犬胆管损伤后胆管狭窄。

【Abstract】 [Objective]To research the relation of PCNA gene and P53 in radioactive inducing dog biliary muscular cell apoptosis. The dogs were randomly divided into the 103pd radioactive stent group (n =6) and the control group (n =6), cross-sections were stained by hematoxylin erosin for dog biliary tissue. The exemination expression of PCNA in biliary duct tissue was detected by ABC immunohistochemical staining as follows the nucleus brow-yellow after ABC coloration was positive cell. So judge positive rate under microscope and exam apoptosis cell of paraffine-bedding boliary duct tissue section using immue histologicochemical method and the examination expression of P53 in biliary duct tissue using hybridization in situ, the nucleus which was brown-yellow after DAB coloration was positive cell. So positive rate could be judged under microscope. Expression of PCNA was more obvious in the control group than in the 103pd radioactive stent group (P <0.01). The biliary duct smooth muscle cell apoptosis rate was more significantly increased in the 103pd radioactive stent group than in the control group(P <0.01), Expression of P53 and PCNA in biliary duct tissue: the expression of P53 was more obvious in the 103pd radioactive stent group than in the control group (P <0.01). [Conclusions] The expression of P53 was more obvious in the 103pd radioactive stent group than in the control group (P <0.01), but PCNA was reduced (P <0.01). The biliary duct smooth muscle cell apoptosis rate was more significantly increased in the 103pd radioactive stent group than in the control group (P <0.01). This result shows that 103pd radioactive stent can inhibit biliary duct smooth muscle proliferation after biliary duct balloon injury through making smooth muscle cell apoptosis, so can inhibit biliary duct stenosis. The resutle shows that 103pd radioactive stent can inhibit biliary duct smooth muscle proliferation.
