【摘要】 目的该实验旨在探讨MMP-2和TIMP-2在浅表性膀胱癌中的表达特点及其与肿瘤复发及进展的关系。方法选取随访资料完整的因膀胱癌而行的膀胱部分切除的病例61例(1995~1997),行常规病理学检查。采用SP免疫组织化学方法,检测61例膀胱癌组织中的7例正常膀胱黏膜的MMP-2和TIMP-2的表达情况。结果正常膀胱黏膜中,MMP-2和TIMP-2均为阴性表达。在61例膀胱癌中,MMP-2的阳性表达为28例,阳性率为45.9%(28/61),MMP-2的表达主要位于肿瘤细胞的细胞质内;TIMP-2在肿瘤细胞的细胞质及间质中均有表达,阳性率分别为50.8%(31/61)和54.1%(33/61)。MMP-2和TIMP-2的表达和肿瘤细胞的病理分级的高低无相关性;TIMP-2间质表达阳性组中浅表性膀胱癌的2年复发率明显高于表达阳性组;在已复发的33例膀胱癌中,MMP-2表达阳性组中进展成为浸润性癌的比率明显高于表达阴性组。结论TIMP-2在间质中的表达可作为浅表性膀胱癌复发的预后指标。MMP-2表达阳性的浅表性膀胱癌在复发过程中易进展为浸润性癌。更多还原

【Abstract】 To study expression of matric metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in superficial carcinoma of bladder and their relationship to tumor recrudescence and progression. We used innumohistochemical method (SP method) to measure levels of the matrix metalloproteinase (TIMP-2) and tissue inhibitor of metalloparoteinase-2 (TIMP-2) in tumor samples from 61 patients (from 1995 to 1997) with bladder cancer treated by cystectomy and in 7 normal specimens of bladder. The expression of MMP-2 and TIMP-2 were negative in normal specimens of bladder the rate of positive expression for MMP-2 was 45.9(28/61) the positive expression of MMP-2 was mostly on cancer cell showed cytoplasmic staining. The positive expression of TIMP-2 was on cancer cell showed in cytoplasm and fibroblast, and the rate of positive expression for TIMP-2 was respectively 50.8%(31/61) and 54.1%(33/61). The presence of MMP-2 and TIMP-2 was not correlated with grade of bladder cancer. In the group of positive expression TIMP-2 in fibroblast, the rate of recurrence of superficial bladder cancer was higher than the group of negative expression. In the patients that had recurred, the rate of progression in the group of positive expression of MMP-2 was higher than the group of negative expression. [Conclusions] The positive expression of TIMP-2 would be indicator of determining the recurrence of superficial bladder cancer. The positive expression of MMP-2 would be indicator of determining the progression of superficial bladder cancer.更多还原
