中药益气解毒颗粒下调端粒酶活性抑制鼻咽癌细胞的生长

【摘要】 目的研究益气解毒颗粒对鼻咽癌细胞端粒酶(Telomerase)活性的影响。方法先用MTT法检测益气解毒颗粒杀伤鼻咽癌细胞系(HNE1)的半抑制浓度(IC50),再用10×IC50,1×IC50,0.5×IC50作用HNE1 12,24,48,72和96 h,然后检测HNE1端粒酶活性;将HNE1接种至BABL/C裸鼠,制备鼻咽癌细胞系裸鼠移植瘤,灌喂中药益气解毒颗粒药液,观察其在体内对鼻咽癌细胞端粒酶活性的影响。结果益气解毒颗粒体外杀伤鼻咽癌细胞的IC50为6.625 mg/mL,10×IC50作用HNE1端粒酶活性下降为阴性(<0.20);1×IC50作用后,端粒酶活性(0.47±0.11)稍有下调,0.1×IC50作用后,端粒酶活性(0.78±0.17),低于空白对照组(1.20±0.23)。灌喂益气解毒颗粒组移植瘤生长受到明显的抑制,抑瘤率为(83.20±7.56)%,端粒酶活性为(0.37±0.10),明显低于对照组(0.89±0.27)。结论益气解毒颗粒在体外和体内均能抑制端粒酶的活性,可能是通过对鼻咽癌细胞端粒酶的活性抑制而发挥抑瘤效应。

【Abstract】 To investigate the inhibitory effect of YIQIJIEDU granule on the telomerase in nasopharyngeal carcinoma. The IC50 of YIQIJIEDU granule on the nasopharyngeal carcinoma cell was detected by using MTT assay, and 10×IC50, 1×IC50, 0.5×IC50 were used to treat HNE1 for 12, 24, 48, 72, 96 h, in which the telomerase activity was detected. 20 nude mice were injected nasopharyngeal carcinoma cells (HNE1), 5×106 cells for a nude mouse. Implanted-tumors grew for 20 d, whose volume reached to 1 cm×1 cm×1 cm. These nude mice were divided into two groups, YIQIJIEDU group and control group. The YIQIJIEDU group was bred YIQIJIEDU granule, and the control was bred normal saline for 30 d. The telomerase activity in implanted-tumors was measured. The IC50 of YIQIJIEDU granule was IC50 6.625 mg/mL on nasopharyngeal carcinoma cells.The telomerase activity was negative in HNE1 treated by 10×IC50 IQIJIEDU granule(<0.20), and it was down-regulated by 1×IC50, which was 0.47±0.10 and was 0.78±0.17 by 0.1×IC50, which was lower than the blank control?The growth of implanted-tumors was inhibited in YIQIJIEDU group, inhibitory rate was 83.2±7.56%,and the telomerase activity was 0.37±0.10, which was lower than the control(0.89±0.27). [Conclusion] IQIJIEDU granule can inhibit the telomerase activity in nasopharyngeal carcinoma in vivo and intro, which it may inhibit nasopharyngeal carcinoma via.
