拟南芥AtNHX5基因的cDNA克隆及其正义、反义表达载体的构建

【摘要】 采用RT-PCR方法从拟南芥品种Landsberg的实生苗叶片中克隆AtNHX5基因的cDNA,核酸序列表明该基因编码区为1554bp,编码538个氨基酸。与GenBank发表的序列(AF490589)相比,有三处碱基不同,分别为850bp处的G(GenBank的为A)、904bp处的T(A)、985处的G(A)。将该基因分别正向或反向置于CaMV35S启动子之后,可构建正义、反义表达载体。

【Abstract】 AtNHX5 gene from Arabidopsis thaliana (Landsberg) was amplified by RT-PCR, and the coding sequence of the gene was analyzed. The result demonstrated that the coding region of AtNHX5 is 1 554 bp, encodes a protein consisting of 538 amino acids. Compared with the sequence in GenBank, there are three different bases from the gene AF490589. In order to study the function of this gene, binary vectors were constructed, in which the coding region of the gene was placed under the 35S promoter in either sense or anti- sense orientation.
