壳寡糖诱导的烟草SKP1基因表达

【摘要】 壳寡糖是一种高效的植物抗性诱导剂,应用mRNA差异显示技术从经过壳寡糖诱导的枯斑三生烟草叶片中分离到了编号为5、31、37和46的4个基因片段。4个基因片段与本塞姆氏烟草(Nicotianabenthamiana)SKP1基因的mRNA同源性都达到82%,据此推断这4个片段是感病烟草品种枯斑三生烟草的SKP1基因。质粒双酶切及反向Northern分析结果表明,该基因表达在壳寡糖诱导下增强。由于SKP1基因与植物的抗病毒病相关,从而在mRNA水平上验证了壳寡糖的诱导抗性效应。

【Abstract】 Oligochitosan is an effective induc-tor for plant resistance. To understand the in-duced resistance mechanism, mRNA differen-tial display was used to isolate genes from Nic-otiana tabacum var. Samsun NN and four en-hanced-expression gene fragments were obtained(Fig.1) and were reamplified. The reamplifiedproducts of appropriate size were isolated andpurified before they were subcloned intoPMD18-T vector. The results of plasmids diges-tion by EcoRI and HindIII and analysis of re-verse Northern blot (Fig.2) indicated that theexpression of the four genes was enhanced byoligochitosan induction. Sequence(Fig.3) and ho-mology analysis show that they share 82% iden- tity in nucleotide sequence with Nicotianabenthamiana SKP1 gene. Because the SKP1 pro-tein as the core component of SCF (ubiquitin li-gase enzyme E3) is relevant to plant resistanceto virus, so these results suggested thatoligochitosan can induce plant resistance and itsmechanism may be relevant to ubiquitination.
