共振光散射法检测水中氨

【摘要】 目的:建立一种测定氨的新方法。方法:人血清白蛋白(HSA)与氨结合,用共振瑞利散射光法进行氨的检测。结果:在368nm处共振光散射增强强度与氨的浓度呈线性关系。以K2HPO4 -NaH2PO4 为缓冲体系维持氨测定溶液的pH=5 80,当温度为309 .16K(32℃) 和静置时间为30min时,方法的检出限为16 4ng/ml,线性范围为0 ~31 5μg/ml,线性相关系数r=0 .994,RSD和回收率分别为1 77% ~5. 35% 和91 .05% ~108 .76%。结论:方法可靠、简便,快速,可直接测定水样中的氨。

【Abstract】 Objctive:To set up a new method for determination of ammonia in water. Methods:The ammonia in water could be determined by resonance light scattering(RLS) method after it has combined with human serum albumin(HSA).Results:The enhanced intensity of RLS at the wavelength 368 nm was proportional to the concentration of ammonia. The buffer solution K 2HPO 4-NaH 2PO 4 was used to maintain the solution pH 5.80, When the temperature and place time were 309.16 K(32℃) and 30 minutes,respectively, the linear range of the calibration curve was 0~ 31.5 μg/ml with the detection limit of 16.4 ng/ml and the correlation coefficient was 0.994.RSD and the rates of recoveries were 1.77%~5.35%,91.05% ~ 108.76% respectively.Conclusion:This method is reliable and accurate and can be used to detect ammonia directly in water.
