血清酮体总量(D-3羟丁酸+乙酰乙酸)测定方法学研究
The study of dtermination method for the total acetone bodies of serum(D-3Hydroxy Butyrate+Cetoacetic Acid)
【摘要】 目的建立一种血清酮体总量(D-3羟丁酸+乙酰乙酸)在全自动生化分析仪上的一种检测方法,用于监测糖尿病酮症酸中毒。方法根据血酮体在体内的代谢过程及体外反应原理,即乙酰乙酸和D-3羟丁酸的相互转化过程,采用酶动力学方法,建立体外定量血清总酮体的测定方法。结果该法操作简便,适用于自动化生化分析仪,各项实验指标达到要求,与英国朗道公司的D-3羟丁酸试剂盒检测的D-3羟丁酸具有高度相关性。结论测定血清酮体总量和单独测D-3羟丁酸相比,可以大大提高酮症的阳性检出率,对及早发现糖尿病酮症,及时纠正酮症及预防酸中毒具有非常重要的临床意义。
【Abstract】 Objective In order to monitor the diabetic ketoacidosis,we managed to establish a method which can used on the full auto-biochemical analysis instrument to determine the total acetone bodies of serum. Methods According to the metabolize process and the reaction theory of the acetone bodies in the serum, we managed to use the enzymatic dynamics method to establish a quantitative measuration to discuss the relative between the total acetone bodies of serum and diabetes mellitus ketoacidosis. Results The method is easy to operate. It can be applicable for the full auto-biochemical analysis instrument. The laboratory indexes are all up to scratch. Its result has the high relativity with the D-3-hydroxy butyrate kit of Randox Laboratories Ltd. of United Kingdom. Conclusion Compared with the determine of D-3-hydroxy butyrate, the determine of the total acetone bodies can increase the positive examination rate of ketoacidosis. It has important significations for the doctors to find the DM ketoacidosis and protect the patient from the DM ketoacidosis as early as possible.
