DPIV酶活性及抗CD26/DPIV抗体与原发性干燥综合征的相关性研究
The study about the correlation of the DPIV enzyme activity and anti-CD26/DPIV antibody with Sjgren’s syndrome
【摘要】 目的探讨二肽蛋白酶(dipeptidylpeptidaseIV)活性及抗CD26/DPIV抗体与原发性干燥综合征之间的相关性。方法用ELISA法检测原发性干燥综合征患者血清中的DPIV酶活性以及抗CD26/DPIV抗体水平,用流式细胞技术检测CD26分子在T细胞上的表达;用免疫印迹法检测人血清中抗CD26/DPIV抗体的存在。结果原发性干燥综合征患者血清中的DPIV酶活性低于健康人;患者体内存在抗CD26/DPIV抗体,并且与DPIV酶活性呈负相关;患者的抗SSB抗体水平与DPIV酶活性呈负相关;79%的人活化T细胞表达CD26分子;患者体内的抗CD26/DPIV抗体水平与健康人体内的无差别,但患者之间的抗体水平有差别,而健康人之间的抗体水平则无差别。结论原发性干燥综合征患者体内产生的抗CD26/DPIV抗体,抗SSB抗体引起DPIV酶活性的下降,可能是原发性干燥综合征重要的发病机制之一;建立了检测DPIV酶活性及抗CD26/DPIV抗体的实验诊断检测方法,对原发性干燥综合征的诊断,鉴别诊断和预后判断有一定的价值。
【Abstract】 Objective To study the correlation of the DPIV enzyme activity and anti-CD26/DPIV antibody with Primary Sjgren syndrome. Methods Flow cytometer assay were adopted to analyze the expression of CD26 on the activated human T cell, and ELISA was used to assay the DPIV enzyme activity and the serum level of anti-CD26/DPIV antibody, and to prove the presence of anti-CD26/DPIV antibody by western-blotting method. Results The specific activity of DPIV enzyme decreased in the sera of the Primary Sjgren’s syndrome patients, and anti-CD26/DPIV antibody didn’t presence only in the sera of patients, but also in the sera of normal human; moreover anti-SSB antibody and anti-CD26/DPIV antibody was associated with the DPIV enzyme activity. Conclusion The decrease of DPIV enzyme activity in the sera of Primary Sjgren syndrome patients could be associated with the production of anti-SSB antibody and anti-CD26/DPIV antibody, it may be involved in the pathophysiology of Primary Sjgren’s syndrome and appeared to be useful as a new disease activity measure of Primary Sjgren’s syndrome.
