DPIV酶活性及抗CD26/DPIV抗体与原发性干燥综合征的相关性研究

【摘要】 目的探讨二肽蛋白酶(dipeptidylpeptidaseIV)活性及抗CD26/DPIV抗体与原发性干燥综合征之间的相关性。方法用ELISA法检测原发性干燥综合征患者血清中的DPIV酶活性以及抗CD26/DPIV抗体水平,用流式细胞技术检测CD26分子在T细胞上的表达;用免疫印迹法检测人血清中抗CD26/DPIV抗体的存在。结果原发性干燥综合征患者血清中的DPIV酶活性低于健康人;患者体内存在抗CD26/DPIV抗体,并且与DPIV酶活性呈负相关;患者的抗SSB抗体水平与DPIV酶活性呈负相关;79%的人活化T细胞表达CD26分子;患者体内的抗CD26/DPIV抗体水平与健康人体内的无差别,但患者之间的抗体水平有差别,而健康人之间的抗体水平则无差别。结论原发性干燥综合征患者体内产生的抗CD26/DPIV抗体,抗SSB抗体引起DPIV酶活性的下降,可能是原发性干燥综合征重要的发病机制之一;建立了检测DPIV酶活性及抗CD26/DPIV抗体的实验诊断检测方法,对原发性干燥综合征的诊断,鉴别诊断和预后判断有一定的价值。

【Abstract】 Objective To study the correlation of the DPIV enzyme activity and anti-CD26/DPIV antibody with Primary Sjgren syndrome. Methods Flow cytometer assay were adopted to analyze the expression of CD26 on the activated human T cell, and ELISA was used to assay the DPIV enzyme activity and the serum level of anti-CD26/DPIV antibody, and to prove the presence of anti-CD26/DPIV antibody by western-blotting method. Results The specific activity of DPIV enzyme decreased in the sera of the Primary Sjgren’s syndrome patients, and anti-CD26/DPIV antibody didn’t presence only in the sera of patients, but also in the sera of normal human; moreover anti-SSB antibody and anti-CD26/DPIV antibody was associated with the DPIV enzyme activity. Conclusion The decrease of DPIV enzyme activity in the sera of Primary Sjgren syndrome patients could be associated with the production of anti-SSB antibody and anti-CD26/DPIV antibody, it may be involved in the pathophysiology of Primary Sjgren’s syndrome and appeared to be useful as a new disease activity measure of Primary Sjgren’s syndrome.
