猪瘟病毒E2糖蛋白A1抗原亚区的表达
Expression of antigenic subdomain A1 of glycoprotein E2 of classical swine fever virus
【摘要】 猪瘟病毒(CSFV)囊膜结构糖蛋白E2(gp55)是诱导机体产生中和抗体及激发保护性免疫应答的主要抗原蛋白。E2存在4个不同抗原区(A-D),根据中和特性和保守性差异,将A抗原区进一步划分为A1、A2、A3抗原亚区,A1抗原亚区在不同猪瘟病毒分离毒株中高度保守且诱导的单克隆抗体具有中和特性。本文采用pRSETC载体,对A抗原区2744nt~2947nt(791aa~858aa)基因片段(EC)进行克隆、表达,并分析表达产物的免疫反应性。研究发现:EC表达产物能与抗猪瘟病毒Alfort毒株E2蛋白的中和性单克隆抗体c2410发生特异性结合,且此结合能被猪瘟病毒抗原或阳性血清阻断。结果表明:A1抗原亚区位于E2蛋白791~858位氨基酸区域。
【Abstract】 Structural and envelope glycoprotein E2 (gp55) of classical swine fever virus (CSFV) is the most antigenic protein being responsible for eliciting neutralizing antibodies and conferring protective immunity. Four antigenic domains, A to D, have been identified on E2. On the basis of neutralization and conservation, domain A is divided into subdomains A1, A2, and A3. Subdomain A1 is conserved in different CSFV strains and the MAbs of the A1 are neutralizing. In this paper, 2 744 nt~2 947 nt( 791 aa~858 aa) gene fragment (EC) of antigenic domain A has been cloned and expressed using pRSET C vector. The immunoreactivity of EC expression product has been analysed and the results showed that the expression product can bind to the neutralizing MAb c2410 (raised against CSFV strain Alfort and reacted with the E2 protein) with specificity, and this binding reactivity can be inhibited or blocked by CSF viral antigen or CSF positive serum. The results indicate that antigenic subdomain A1 is located between residues 791 and 858 of E2 protein.
