MMP-2及CD147在侵袭性垂体腺瘤中的表达及其相互关系的研究

【摘要】 目的检测基质金属蛋白酶(MMP-2)及CD147在侵袭性和非侵袭性垂体腺瘤中的表达,以探讨其与肿瘤侵袭发生的关系及两者之间的相关性。方法用免疫组化SP法和RTPCR方法对55例垂体腺瘤患者标本MMP2及CD147的表达进行检测。结果免疫组化显示,侵袭性垂体腺瘤的MMP-2强阳性及阳性表达率明显高于非侵袭性垂体腺瘤(65.6%和93.8%,13.0%和43.5%,P<0.05),CD147强阳性及阳性表达率明显高于非侵袭性垂体腺瘤(53.1%和96.9%,21.7%和52.2%,P<0.01)。侵袭性垂体腺瘤组MMP-2及CD147mRNA的表达(1.126±0.081,0.887±0.043,P<0.01),明显高于非侵袭性垂体腺瘤组(0.425±0.083,0.179±0.051,P<0.01)。侵袭性腺瘤组中MMP2与CD147的表达成正相关(r=0.73,P<0.05),非侵袭性腺瘤组中MMP-2与CD147的表达无明显相关性(r=0.34,P>0.05)。结论MMP2及CD147的高表达与垂体腺瘤的侵袭性密切相关。MMP-2和CD147可以作为垂体腺瘤侵袭性的2项有效指标。在肿瘤侵袭过程中CD147与MMP-2的产生关系密切。

【Abstract】 Objective To investigate the expression of matrix metalloproteinase-2(MMP-2) and CD147 in invasive pituitary adenomas,and to evaluate the relationship between MMP-2 and CD147 and the invasion of pituitary adenomas. Methods MMP-2 and CD147 expression were examined by immunohistochemical method, and the expression of MMP-2 and CD147 mRNA were examined by RT-PCR assay. Results The intense expression and total expression rate of MMP-2 in patients with invasived pituitary adenomas invasion(65.6%,93.8% respectively) were significantly higher than those without invasion (13.0%, 43.5% respectively, P<0.05), and the intense expression and total expression rate of CD147 of pituitary adenomas patients with invasion(53.1%, 96.9% respectively) were significantly higher than those without invasion (21.7%,52.2% respectively, P<0.01). Both mRNA expression of MMP-2 and CD147 were also observed significantly higher in invasive pituitary adenomas(1.126±0.081, 0.887±0.043 respectively)than those without invasion(0.425±0.083, 0.179±0.051 respectively, P<0.01).There were positive correlations between MMP-2 and CD147 mRNA expression in invasive pituitary adenomas (r=0.73, P<0.05), and no correlations between them in noninvasive pituitary adenomas(r=0.34, P>0.05). Conclusions The study suggestted that MMP-2 and CD147 might be associated with the aggressiveness and invasion in pituitary adenomas.MMP-2 and CD147 are two useful tools for assessing the invasion of pituitary adenomas. CD147 has closed relationship with the creating of MMP-2 in the invasion of pituitary adenomas.
