大豆异黄酮对去卵巢大鼠子宫细胞凋亡影响的研究
Effects of soybean isoflavone on uterus apoptosis in ovariectomized rats
【摘要】 目的:探讨大豆异黄酮对去卵巢大鼠子宫细胞凋亡的影响。方法:60只雌性Wistar大鼠,随机分为6组, 正常组、模型组、雌二醇组、假手术组、大豆异黄酮低剂量组、大豆异黄酮高剂量组。给药6周后,处死大鼠,分 离摘取子宫,常规石蜡包埋切片。凋亡细胞的检测使用TDT mediateddUTP biotinnickendlabeling(TUNEL)方 法。结果:TUNEL标记率为:正常组粘膜层和肌层分别3.84%和6.26%,模型组为14.18%和17.33%,雌二醇 组为0.88%和0.40%,假手术组为3.74%和5.84%,大豆异黄酮低剂量组为7.78%和4.60%,大豆异黄酮高 剂量组为2.26%和0.92%。与模型组比较各组凋亡率明显减少(P<0.01)。结论:大豆异黄酮能明显抑制去 卵巢大鼠子宫细胞凋亡。
【Abstract】 Objective:To study the effect of soybean isoflavone on uterus apoptosis in ovariectomized rats.Methods:60 Female Wistar rats were randomly divided into 6 groups:normal control group,model group,estrogen group,sham operation group,soybean isoflavone group in low dose and soybean isoflavone group in high dose .Six weeks later,the rats were sacrificed and the isolated uterus were fixed with formalin.The apoptosis rates were measured by TDT mediated dUTP biotin nick end labeling(TUNEL) method.Results:The apoptosis rate of endometrium and myometrium in normal control group were 3.84% and 6.26% respectively,the model group were 14.18% and 17.33%,the estrogen group were 0.88% and 0.40%,the sham operation group were 3.74% and 5.84%, the low dose group was 7.78% and 5.60%, the high dose group was 2.26% and 0.92%.Compared with normal control group,the mortality rates of other groups were significantly reduced( P< 0.01).Conclusion:Soybean isoflavone can restrain the uterus apoptosis in ovariectomized rats.
