用快速分离柱高效液相色谱法测定生物发酵产物中的乳酸
Study on the determination of the lactic acid in fermentation production by HPLC with rapid separation column
【摘要】 对用快速分离柱高效液相色谱法测定生物发酵产物中乳酸的方法进行了研究。以ZORBAX Stable Bound(4.6×50mm,1.8μm)C18快速分离柱为固定相,甲醇和0.01mol/L的磷酸5:95为流动相,流速为2.0mL/min。在该色谱条件下,发酵产物中的乳酸在2.0min内可达到基线分离;用紫外二极管矩阵检测器检测,方法标准回收率为97% ̄102%,相对标准偏差为0.95% ̄1.28%。
【Abstract】 A HPLC with rapid separation column for the determination of lactic acid in fermentation was studied. The lactic acid was separated on a ZORBAX Stable Bound(4.6×50 mm, 1.8 μm) rapid separation column, with methanol - 0.01 mol/L H3PO4 (5:95) as mobile phase at a flow rate of 2.0 ml/min, and monitored with the photodiode array detector at 210 nm. Under such conditions, lactic acid produced in fermentation could be separated at the bottom line. The recovery rate of lactic acid was 97%~102%, and the relative standard deviation was 0.95%~1.28%. The method was applied to measure lactic acid in different fermentation production with satisfied results.
