固态发酵啤酒糟产木聚糖酶的菌种筛选及其产酶条件研究
Study on screening of strain used for producing xylanase from beer grains with solid state fermentation and the condition of producing xylanase
【摘要】 从40株保藏菌种中筛选得到1株能发酵啤酒糟高产木聚糖酶的菌株黑曲霉AN27-2-1。通过单因素实验和正交实验对黑曲霉AN27-2-1固态发酵啤酒糟产木聚糖酶的条件进行优化,结果表明:在培养基组分为啤酒糟:玉米芯:麸皮=6:2:2,NH4NO3 2%,吐温800.1%,加水比为1:2,接种量为3.5%,培养温度为30℃,培养周期为2d~3d,产酶活达310.54U/g。
【Abstract】 A strain Aspergillus niger AN27-2-1, which has ability to highly produce xylanase with beer gains was obtained from 40 preserved strains. The conditions for producing xylanase from beer grains were optimized through single factor experiment and orthogonal experiment and were showed as follows, the ratio of beer grain: corncob: bran in medium 6:2:2; the content of NH4NO3 2%; the content of tween 80 0.1%; the ratio of water to medium 1:2; the inoculum 2.0%; the temperature 30℃; the time for fermentation 2-3 d. Under those conditions, the enzyme activity of xylanase produced by the strain AN27-2-1 reached 310.54 IU/g.
