冠心病患者C-反应蛋白+1444C/T基因多态性的研究

【摘要】 目的探讨中国汉族人群C反应蛋白(CRP)+1444C/T基因多态性与CRP水平及冠心病(CHD)的关系。方法提取128例CHD患者(至少1支冠状动脉直径狭窄≥50%)和119例对照者的基因组DNA,采用PCR限制性片段长度多态性(PCRRFLP)方法检测CRP+1444C/T基因多态性,同时测定血清超敏CRP(hsCRP)及血脂水平。结果湖北地区汉族人群中存在CRP+1444C/T多态性,在冠心病组中基因型分布为:CC型89.1%,CT型10.9%,对照组分别为89.9%和10.1%。CHD组和对照组之间的基因型频率和等位基因频率均无显著性差异(P>0.05),但血清CRP水平在对照组不同基因型之间有显著性差异(P<0.05)。结论CRP+1444C/T基因多态性与正常人CRP水平高度相关,但未发现其与湖北汉族人群CHD发病有关。

【Abstract】 Objective To explore the relationship between C reactive protein (CRP) +1444C/T gene polymorphisms and coronary heart disease (CHD) and assess their effect on CRP levels. Methods CRP +1444C/T gene polymorphism of 128 cases with CHD and 119 normal controls were determined by PCR-restriction fragment length polymorphism (RFLP). Meanwhile serum hypersensitive CRP (hsCRP) and plasma lipid levels were measured in all subjects. Results Han nationality population in Hubei had CRP+1444C/T polymorphisms, the frequencies of CC, CT genotypes were 89.1% and 10.9% in CHD group while ones were 89.9% and 10.1% in controls. There were no significant differences in frequencies of genotype and allele between controls and CHD cases (P>0.05). Conclusions (CRP+)1444C/T polymorphisms should be related to CRP levels in normal individuals, but be independent of occurrence of CHD in Han nationality population in Hubei.
