大鼠局灶脑缺血预处理对caspase-3表达影响的研究

【摘要】 目的观察对比caspase3在大鼠局灶脑缺血预处理和脑缺血再灌注中的表达。方法采用线栓法局灶脑缺血模型,ABC免疫组化方法检测caspase3表达。结果caspase3免疫反应阳性细胞数缺血预处理组(IPC+MCAO)明显低于单纯局灶缺血组(SS+MCAO),阳性细胞呈棕褐色,清晰可辨,IPC+MCAO组caspase3蛋白阳性细胞在缺血6h表达开始增多,24h达高峰,72h下降;SS+MCAO组caspase3蛋白阳性细胞表达时相一致,但增高明显。结论缺血预处理可能通过降低caspase3表达,对缺血引起的神经损伤起到保护作用。

【Abstract】 Objective To investigate and compare the differences in caspase-3 expressions after focal cerebral ischemic preconditioning (IPC) and cerebral ischemic reperfusion in rats. Methods Caspase-3 expressions in Longa’ rat model for focal cerebral ischemia were determined by ABC immunohistochemistry technique. Results The positive caspase-3 immunoreactivity in IPC+MCAO group were fewer than that in SS+MCAO group, presented chocolate brown and identified clearly; that in IPC+MCAO group increased at 6 h after ischemia, reached peak at 24 h, reduced at 72 h; that of SS+MCAO group was same with significant increase. Conclusions Focal ischemic preconditioning might be play a protective role in neurological impairment induced by ischemia through decreasing caspase-3 expressions.
