同种异体移植瘤在不同组织微环境中生物学行为的差异及机制

【摘要】 目的 分析不同的组织微环境对同种异体移植瘤生物学行为的影响因素,并探讨基质金属蛋白酶和转化生长因子β1 与移植瘤生物学行为的关系。方法 应用常规HE染色观察不同移植瘤的生物学行为表现,并应用免疫组化的方法分别检测两种瘤组织中MMP 2,MMP 9 及TGF β1 的表达情况。结果 ①不同的组织微环境同种异体移植瘤生物学行为有明显差别,皮下转移瘤表现为明显的膨胀性生长;腹腔转移瘤表现为显著的浸润性生长;②MMP 2 及MMP 9 的表达:皮下转移瘤两者均为阴性;腹腔转移瘤可见MMP 2及MMP 9阳性表达;③TGF β1 的表达:皮下转移瘤可见TGF β1 的间质型表达;腹腔转移瘤可见TGF β1 的胞浆型表达。结论 不同组织环境对同种移植瘤的浸润及转移有影响,MMP 2、MMP 9及TGF 表达可能是其中的因素之一。

【Abstract】 Objective To study the effect elements about different organic micro-environment to and investigate the relationship between MMPs, TGF-β 1 and transplant-tumor’s biological deed. Methods The staining with HE method is conducted to investigate the presentation of different transplant-tumor’s biological deed. Immunohistochemistry is used to detect the expression of MMP-2, MMP-9, TGF-β 1 in two tumor organ respectively. Results ①The biological deeds of identical transplant-tumor′s from different body have significant difference in different organic micor-envionment. subcutaneous metastaic carcinoma presents expensive grouth; Belly metastaic carcinoma presents infiltrative grouth. ②The expression of MMP-2 and MMP-9: Subcutaneous metastaic carcinoma is negative; belly metastaic carcinoma is positive. ③The expression of TGF-β 1: In subcutaneons metastaic carcinoma, TGFβ 1 is expressed in interstitial tissue. In belly metastaic carcinoma, TGF-β 1 is expressed in cytoplasm. Conclusion Different organicmicro-environment will affect the metastasis and infiltration of identical transplant-tumor. The expression of MMP-2, MMP-9 and TGFβ 1 is one of its elements.
