头孢地嗪对中性粒细胞细胞因子分泌的影响

【摘要】 目的研究头孢地嗪(cefod iz im e,CDZ)对肺炎克雷伯菌(K lebsiella pneum on iae)感染诱导的小鼠中性粒细胞因子分泌的影响,并初步探讨其与LPS-TLR-NFκB信号传导通路的关系。方法用RT-PCR、EL ISA等方法检测肺炎克雷伯菌感染诱导中性粒细胞分泌的TNFα-、IL-1β改变和T o ll like receptor 4(TLR 4)的表达;W estern b lotting检测I-κB活性,以及CDZ对上述指标的影响。结果CDZ促进了肺炎克雷伯菌感染早期小鼠中性粒细胞TNFα-、IL-1β分泌及TLR 4的mRNA表达,同时伴有I-κB的降解增强;在感染后期CDZ降低了中性粒细胞TNFα-、IL-1β的分泌,但仍可促进TLR 4表达。结论CDZ对肺炎克雷伯菌感染诱导的小鼠中性粒细胞有免疫调节作用,此作用可能与其调节TLR 4表达和I-κB活性而影响TNFα-、IL-1β等促炎因子基因转录有关。

【Abstract】 Objective To explore the immunoregulating effects of cefodizime(CDZ) on cytokines production of neutrophil in response to heat-killed Klebsiella pneumoniae in mice.Methods The effects of CDZ on cytokines production of neutrophil induced by K.pneumoniae in mice were detected by RT-PCR and ELISA.We also studied the changes of signal transduction pathway in this process by evaluating the expression of the inhibitor factor of κBα(I-κBα) and toll like receptor 4(TLR4) in neutrophil by Western blotting and RT-PCR respectively.Results CDZ treatment promoted the neutrophil cytokines production.CDZ increased(neutrophil) TNF-α,IL-1β production and the degradation of I-κB in the early stage after K.pneumoniae infection in mice.However,in the late stage of infection,CDZ reduced the production of TNF-α and IL-1β and decreased the activities of NF-κB activity in infected mice.CDZ improved mRNA expression of TLR4.Conclusion CDZ could alter neutrophil cytokine production in K.pneumoniae infected mice through LPS-TLR-NFκB pathway by regulating the transcription of TNF-α and IL-1β in nuclear.
