γ-羟基丁酸对缺血/再灌注损伤后神经细胞凋亡及相关基因Bcl-2、Bax表达的影响
Study on the effect of GHBA on the apoptosis of the nervous cell and Bcl-2,Bax protein expression after focal cerebral ischemia/reperfusion injury
【摘要】 目的探讨γ-羟基丁酸(GHBA)对缺血/再灌注神经细胞凋亡及相关基因Bcl-2、Bax表达的影响及其可能的作用机制。方法采用大鼠局灶性缺血/再灌注损伤模型,大脑中动脉阻塞1h,分别于再灌注后1.5、3、6、12、24、48、72h,用原位末端缺口标记法(TUNEL)和免疫组织化学方法检测假手术组、生理盐水组和GHBA组海马区的凋亡细胞、Bcl-2、Bax的表达。结果与生理盐水组比较,GHBA组细胞凋亡显著减少(P<0.05),Bcl-2表达增多,Bax表达减少,且Bcl-2/Bax的比率上调(P<0.05)。结论GHBA可能通过增加Bcl-2表达、抑制Bax表达,抑制神经细胞凋亡,并可影响Bcl-2、Bax的平衡。
【Abstract】 Objective To observe the effect and possible mechanism of GHBA on the apoptosis of the nervous cell and Bcl-2, Bax protein expression after focal cerebral ischemia/reperfusion injury in the rats. Methods Focal cerebral ischemia/reperfusion injury models were prepared by 1 hours of occlusion followed by 1.5,3,6,12,24,48,72 hours of reperfusion of the middle cerebral arteries (MCA) in rats. TUNEL and immunohistochemical technique were used in hippocampal area to examine the apoptotic cell and the expressions of Bcl-2,Bax protein in the sham-operation group, the NaCl group and the GHBA group,respectively. Results ①The number of TUNEL positive cell in GHBA group was larger than that in NaCl group(P<0.05).②The expression of Bcl - 2 was more and the expression of Bax was less in hippocampal area in GHBA group than those in NaCl group (P<0. 05), and GHBA can increase the ratio of Bcl-2/Bax. Conclusion GHBA may protect cerebral neurons and inhibite apoptosis by increasing the expression of Bcl-2 and inhibiting the expression of Bax,and may influence the balance between Bcl-2 and Bax.
