茶汁中灵芝菌产多酚氧化酶发酵条件的研究
Studies on fermentation conditions of Ganoderma lucidum producing polyphenol oxidase in the low-grade green tea
【摘要】 绿茶汁中茶多酚一方面对灵芝菌菌体生长有一定的抑制作用,另一方面,低浓度的茶多酚能诱导灵芝菌分泌多酚氧化酶.多酚氧化酶可降解发酵液中多酚类物质,减少酚类物质对菌体生长的影响,积累更多的胞外代谢产物,降低发酵液酚氨比,提高茶色素含量,因此在低档绿茶汁中研究灵芝菌深层液体发酵产多酚氧化酶,对进一步研究发酵代谢控制具有重要意义.试验采用正交设计考察不同发酵培养基组分对灵芝菌在茶汁中产多酚氧化酶的影响情况,结果表明:最佳发酵培养基组合为蔗糖2.5%,蛋白胨0.25%,茶汁2.5%,KH2PO40.20%;该菌的最适发酵工艺条件为发酵液pH5,摇床转速200r.mi-n1,培养温度30℃,接种量10%,装液量20%,培养时间为7d,其多酚氧化酶活性达到95U.mL-1.
【Abstract】 Tea polyphenol (TP) in the low-grade green tea could inhibit Ganoderma lucidum to grow, but the low concentration TP might induce Ganoderma lucidum to produce polyphenol oxidase (PPO). When PPO decomposed the polyphenol during fermentation process, the effect of the polyphenol on Ganoderma lucidum growth was reduced, extracellular metabolic products were accumulated, the ratio of polyphenol and amino acid was declined and the concentration of tea pigment was raised. Consequently, it was of great importance to study the metabolic control further. In this work, using orthogonal test, the effect of different compositions of fermentation medium on Ganoderma lucidum producing PPO in the low-grade green tea was evaluated and the best medium compositions were as follow: sucrose 2.5%, peptone0.25%, low-grade green tea2.5%, KH2PO40.2%. It was also found that the best fermention coditions were pH 5.0, 200 rmin-1, 30 , inoculum volume 10%, cultivating liquor 20%, cultivating time 7 d, and in these optimized conditions the enzyme activity might arrive at 95 UmL-1.
