【摘要】 目的探讨Ⅰ、Ⅲ型前胶原末端肽在评价试验性高血压大鼠(SHR)颈动脉血管间质胶原重构及逆转中的意义。方法选择16周龄雄性SHR各12只分别为SHR组和厄贝沙坦组(30mg·kg-1·d-1),另选同龄雄性Wistar大鼠12只为正常对照组;颈总动脉切片采用HE染色、苦味酸———天狼星红(PSR)染色分别检测颈动脉壁厚/腔径比值、胶原面积百分比;采用放免法测定三组血清中PCⅠ、PCⅢ的浓度。结果SHR组颈动脉壁厚/腔径比值较正常对照组高,厄贝沙坦组治疗20周后,较SHR组下降;SHR组胶原面积百分比较正常对照组高,厄贝沙坦组治疗20周后,较SHR组也有下降;SHR组血清PCⅠ、PCⅢ和PCⅠ/Ⅲ均较正常对照组明显增高,应用厄贝沙坦后PCⅢ下降明显,与正常对照组的差异亦无意义;PCⅠ和PCⅠ/Ⅲ虽有下降,但与正常对照组的差距仍然明显。结论SHR颈动脉血管存在间质胶原重构,应用厄贝沙坦可以得到一定程度的逆转,血清PCⅠ能同步反映血管胶原面积百分比的变化,能够用于评价试验性高血压大鼠颈动脉血管胶原重构及逆转的变化。更多还原

【Abstract】 Objective To investigate the ch ange of serum concentrations of procollagen type Ⅰ and Ⅲ in SHR and its signif icance in carotid artery vascular remodeling.Methods 12 SHR tre ated with irbesartan,12 SHR without treated irbesartan,12 Wistar rats were norma l control group.The vascular volume fraction of collagen and the ratio of wall/c avity were determined by picrosirius red and HE staining.The serum concentration s of procollagen type Ⅰ(PCⅠ) and procollagen type Ⅲ(PCⅢ) were measured by sp ecific radioimmunoassays.Results Significant fibrosis exists in carotid artery of SHR.After 20 weeks’ treating,the vascular volume fraction of collagen and the ratio of wall/cavity were descended.Baseline serum PCⅠ,PCⅢ an d the ratio of PCⅠ/Ⅲ were increased in SHR.The correlation between PCⅠ with v olume fraction of collagen was positive,PCⅢ and the ratio of PCⅠ/Ⅲ with volum e fraction of collagen were no significant correlation in the SHR.Conclu sion The serum concentration of PCⅠ can indicate the significance of t he carotid artery vascular collagen remodeling in SHR.更多还原
