氯喹治疗小鼠弓形虫感染的实验观察
EXPERIMENTAL OBSERVATIONS OF CHLOROQUINE IN THE TREATMENT OF TOXOPLASMA GONDII INFECTION IN MICE
【摘要】 目的研究和探索治疗弓形虫病新的有效药物,并揭示其治疗机理。方法用弓形虫Rh株感染昆明小鼠,设计氯喹治疗组、甲硝唑组及对照组,观察对比。扫描电镜及透射电镜下观察对比各种情况下弓形虫的超微结构变化。结果按照疗效判定标准,氯喹治疗小鼠弓形虫感染有效,而甲硝唑无效。扫描电镜观察发现,氯喹治疗组中弓形虫速殖子有聚集、粘连,虫体大小不均,表面皱缩,形态不规则。透射电镜观察对照组大量有核细胞内有弓形虫侵入并增殖,以一个细胞内3~4个弓形虫速殖子者多见。氯喹治疗组中少数组织细胞内有弓形虫侵入,且细胞内仅有1个弓形虫速殖子;大部分弓形虫细胞膜及核膜均有破损,破膜从头、尾两端开始;弓形虫内脂肪空泡及线粒体数量较对照组明显减少,棒状体断裂。结论氯喹是一种非常有效的治疗小鼠弓形虫病药物。其治疗机理是破坏弓形虫的细胞膜及核膜,降低其攻击有核细胞的能力,阻断其在细胞内的增殖,且造成代谢紊乱。
【Abstract】 Objective To explore a new and effective drug for the treatment of toxoplasamosis, and reveal its therapeutic mechanism. Methods Mice were infected with RH Toxoplasma gondii, and divided into chloroquinetreatment group: metronidazolumtreatment group and control group. Scanning electron microscope(SEM) and transmission electron microscope(TEM) were used to observe the ultramicrostructural changes in T. gondii under all kinds of conditions mentioned above. Results According to the treatment criteria, it was shown that chloroquine was effective in the treatment of toxoplasamosis, but metronidazolum was not. In SEM, it was found that the tachyzoites of T. gondii began to come together and adhered with each other, the sizes appeared uneven and the surface became shrunk and irregular. In the observations with TEM, it was found that in the control group there were invasions and multiplications of tachyzoites in a large number of nucleated cells. Usually 3-4 tachyzoites can be seen in one cell; In the choloroquinetreatment group, only very few cells were invaded and usually with one tachyzoite. The membrane and nucleus of most tachyzoites were damaged and the membrane damages occurred from the both ends. The number of fat vacuoles in the tachyzoites dropped sharply. Conclusion It was suggested that chloroquine can be an effective drug in the treatment of toxoplasamosis. The mechanism of it is likely that chloroquine can weaken its ability to invade into the nucleated cells and stop its own multiplications by damaging the membrane and nucleus of tachyzoites. At the same time, the metabolism of lipid in the tachyzoites became abnormal.
