乳腺癌患者外周血CD4+CD25+调节性T细胞的检测及意义

【摘要】 目的探讨乳腺癌患者外周血中CD4+CD25+调节性T细胞的变化及意义。方法采用流式细胞技术检测64例乳腺癌患者外周血中CD4+CD25+调节性T细胞,采用ELISA法检测外周血中转化生长因子β1(TGFβ1)的表达水平。结果乳腺癌患者外周血中CD4+CD25+调节性T细胞占T淋巴细胞的百分比为(5.1±2.9)%,高于乳腺良性肿物患者和健康志愿者(P均<0.05)。乳腺癌患者外周血中CD4+CD25+T细胞水平与肿物大小、TGFβ1呈正相关(r分别为0.511和0.253),与CD8+CD28+T细胞和NK细胞呈负相关(r分别为-0.243和-0.301)。结论乳腺癌患者外周血中具有免疫抑制活性的CD4+CD25+调节性T细胞水平较高,对乳腺癌患者具有免疫抑制作用。

【Abstract】 Objective To evaluate the changes of CD4+CD 25 + T cells in peripheral blood from patients with breast cancer. Methods Sixty four patients with breast cancer, 15 patients with benign breast tumors and 9 healthy volunteers were included in this study. The proportion of CD4+CD 25 + T cells population in total T cells was evaluated by flow cytometric analysis. The cytokine production(TGF-β1)was measured by ELISA. Results The population of CD4+CD 25 + T cells in peripheral blood from patients with breast cancer accounted for (5.1± 2.9)% of the total amount of T lymphocytes, and was significantly higher in comparison with that in patients with benign tumors and in healthy volunteers (P<0.05). The CD4+CD 25 + T cells population in breast cancer patients was positively correlated with the cancer size and with TGF-β1 level (r=0.511 and r=0.253, respectively), and negatively correlated with CD8+CD 28 + T cells and NK cells (r =-0.243 and r=- 0.301 , respectively). Conclusion The CD4+CD 25 + regulatory T cells in peripheral blood of patients with breast cancer is significantly increased in comparison with that in patients with benign breast tumor and in healthy subjects. It may be responsible for immune suppression in breast cancer patients.
