载脂蛋白E基因多态性与新疆维吾尔族自然长寿的相关性研究

【摘要】 目的探讨载脂蛋白E(apolipoproteinE,apoE)基因多态性与新疆维吾尔族自然长寿的关系。方法应用聚合酶链反应限制性片段长度多态性方法检测百岁组42名,90岁组102名,65~70岁组70名和对照组53名的apoE基因多态性。结果百岁组apoE的ε3/3、ε2/3和ε3/4基因型频率分别为69.0%、23.8%和2.4%,其ε3、ε2和ε4等位基因频率分别为82.1%、16.7%和1.2%,百岁组ε3/4基因型及ε4、ε3等位基因频率显著低于对照组(P<0.01),ε2/3基因型及ε2等位基因频率则显著高于对照组(P<0.01)。百岁与apoE基因的ε2等位基因呈正关联,与ε4等位基因呈负关联。结论在新疆维吾尔族,apoE基因多态性与个体寿命密切相关,同时也应考虑到长寿是年龄依赖的多种因素影响的结果。

【Abstract】 Objective To explore the distribution of polymorphisms of apolipoprotein E(apoE) gene and its relation to natural longevity in the Xinjiang Uighur people. Methods Using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP), the authors obtained data from centenarians(n=42), 90-year-old people (n=102), 65-70-year-old people (n=70) and controls(n=53). Results The frequencies of apoE genotypes ε3/4,ε3/3 and ε2/3 were 2.4%,69.0% and 23.8% respectively in the centenarian group, and the frequencies of their ε4,ε3 and ε2 alleles were 1.2%,82.1% and 16.7%. The frequencies of genotype ε3/4 and ε4,ε3 allele in the centenarian group were significantly lower than those in controls(P<0.01), whereas the frequencies of genotype ε2/3 and ε2 allele in the centenarian group were significantly higher than those in controls(P<0.01). Longevity was noted to be positively correlated with ε2 and negatively correlated with ε4. Conclusion This relationship reflects a genetic influence on differential survival and may point to pleiotropic age-dependent effects on longevity.
