【摘要】 目的探讨组织工程方法构建血管补片的可行性。方法用壳聚糖和透明质酸多孔多聚体支架构建高分子复合材料支架,将犬的骨髓细胞种植其中,并植入自身犬的肺动脉上,观察材料上细胞外基质生成及表面内皮化的情况,分别在术后2周、4周、8周用电镜、组化等方法来进行评价。结果所有动物在实验期间均存活,组织学及免疫组织化学显示,术后补片形成类似血管壁样组织。结论此复合材料和骨髓细胞可用来构建组织工程血管补片,其结构与血管壁类似。

【Abstract】 Objective To investigate the possibility of constructing vascular patches by means of tissue engineering. Methods The biodegradable scaffolds were implantated subcutaneously to nude mice and the mice were sacrificed in 4 and 8 weeks, respectively . Cell-biodegradable scaffold mixtures were made by separately seeding cells from canine bone marrow onto biodegradable scaffold and implanted into the pulmonary artery of dogs. The implanted grafts were analyzed at 2,4, and 8 weeks after implantation by histology, immunohistochernistry, and electron microscope. Results All animals survived the procedure. The tissue-engineered vascular patches were patent with no sign of thrombus formation. The gross appearance of patches was similar to that of the natural counterparts. Histologic and immunohistochemical analyses of the neoformed tissues revealed typical formation of organized and functional tissue. Conclusion Synthetic polymer vascular patches with histology similar to natural vessel can be constructed by tissue engineering.
