一种经酶处理新型动脉瘤动物模型的建立
Construction of a new aneurysmal models
【摘要】 目的建立与临床动脉瘤更加相似的动脉瘤动物模型。方法利用手术吻合法将15只大白兔制成15个顶端动脉瘤模型。制作后当日及2周后行形态、病理和行为表现的观察。采用自行设计循环加压装置,显微镜下使用高速摄影机记录试件整体形态的变化。通过图形分析软件测量试件变化值。结果共制作顶端动脉瘤模型15个,1个术后24h破裂,1个2周后破裂。动脉瘤模型建立后的平均直径为(2.10±0.24)mm(n=15),2周后动脉瘤平均直径为(3.25±0.54)mm(n=13),两者差异有统计学意义(P<0.05)。2周后病理检查可见瘤壁弹力层和胶原纤维明显减少或消失。得出试件的材料常数x1,x2均值分别为4.25602、113.0667和655.1316,0.2566292。构建出动脉瘤壁的本构方程σ=x1x2λ2Eexp[E2-E2)]。结论经酶处理的动脉瘤动物模型形态和病理上与临床动脉瘤更加相似。
【Abstract】 Objective To develop aneurysmal models which are similar to clinical counterparts.Methods Fifteen rabbit’s terminal aneurismal models were made by transplantration.The diameters were measured just after the construction and 2 weeks after the construction of the models.The aneurismal models and their parent arteries were resected for the research of modality,pathology and behavior.In the circle equipment of exerting press,the model was recorded by high-speed video camera under the microscopy.The changing numerical value was measured with the figure analyzing software.[WT5”HZ]Results Fifteen aneurismal models were successfully prepared.One model ruptured 24 h after the construction and one ruptured 2 weeks after the construction.The aneurismal models were enlarged from the initial mean diameters of (2.10± 0.24) mm (n=15) to (3.25± 0.54) mm (n=13) 2 weeks after the construction.There was significant difference between the two groups in diameters.Pathological examination of the experimental aneurysms revealed that layers of elastic fibers and collegen were obviously decreased or vanished.The material constant of two groups was x1 (4.256 02 and 655.131 6) and x2 (113.066 7 and 0.256 629 2) respectively with the difference being significant (P< 0.05).Conclusion The aneurismal models constructed by transplantation of elastase and collagenase digested arterial segments are more similar to clinical counterparts morphologically and pathologically.
