PTEN与hTERT在肝细胞肝癌中的表达及其相关性研究

【摘要】 目的 研究肝细胞肝癌组织中PTEN与hTERT的表达及其意义 ,并探讨两者在肝细胞肝癌中表达的相关性。方法 应用免疫组织化学 (SP法 )和半定量逆转录 聚合酶链反应 (RT PCR)分析 5 8例肝细胞肝癌及对应癌旁组织中PTEN蛋白及PTENmRNA、hTERT蛋白及hTERTmRNA的表达情况 ,并探讨两者表达的相关性。结果 PTEN蛋白明确定位于肝细胞胞浆内。PTENmRNA和PTEN蛋白在肝细胞肝癌中的阳性率分别为 41.3 8% (2 4/ 5 8)、44 .83 % (2 6/ 5 8) ,明显低于癌旁肝组织的阳性率 10 0 % (5 8/ 5 8) ,差异有统计学意义 (P <0 .0 1)。hTERT蛋白主要表达于肝细胞胞浆内 ,偶见核内表达。hTERTmRNA和hTERT蛋白在肝细胞肝癌中的阳性率分别为82 .76% (4 8/ 5 8)和 87.93 % (5 1/ 5 8) ,明显高于癌旁组织中的阳性率 2 4.14 % (14 / 5 8)、2 7.5 9% (16/5 8) ,差异有统计学意义 (P <0 .0 1)。相关分析显示 ,PTEN与hTERT在肝细胞肝癌中的表达呈负相关 (P <0 .0 5 )。结论 PTEN、hTERT在肝细胞肝癌的发生发展中起重要作用 ,有可能成为肝细胞肝癌的重要分子生物学检测指标 ,且两者表达呈负相关 ,提示PTEN可能在肝细胞肝癌的发生发展过程中对hTERT的活化起着调控作用。

【Abstract】 Objective To investigate the significance of the expression of PTEN and hTERT gene and their correlation in human hepatocellular carcinoma (HCC).Methods The expression of PTEN protein and hTERT protein in HCC and their corresponding para carcinoma liver tissues from 58 HCC patients was detected by immunohistochemistry (SP method) and the expression of PTEN mRNA and hTERT mRNA by reverse transcription polymerase chain reaction (RT PCR) respectively.The correlation between PTEN and hTERT gene in HCC was analyzed.Results The expression of PTEN protein was definitely located in cytoplasma of HCC.The positive rate of PTEN mRNA and PTEN protein in HCC was respectively 41.38% (24/58) and 44.83% (26/58),significantly lower than that in the corresponding para carcinoma liver tissues:100% (58/58) ( P < 0.01). The expression of hTERT protein was mostly located in cytoplasma of HCC,occasionally in nucleus.The positive rate of hTERT mRNA and hTERT protein in HCC were respectively 82.76% (48/58) and 87.93% (51/58),obviously higher than that in the corresponding para carcinoma liver tissues:24.14% (14/58) and 27.59% (16/58) respectively ( P < 0.01). There was a negative correlation between PTEN and hTERT gene expression in HCC ( P < 0.05). Conclusion Loss of PTEN gene expression and positive expression of hTERT gene may play an important role in the genesis and development of HCC.PTEN and hTERT gene may serve as an importment molecular biological indicators in diagnosing and predicating the biological behavior of HCC.The significant negative correlation between the expression of PTEN and hTERTgene indicates the one of the approaches of hTERT activation and up regulation may be conferred by the loss of PTEN gene expression in HCC.
