EGFR、PCNA、N-myc和P16基因蛋白产物在髓母细胞瘤中的表达
Expression of EGFR,PCNA,N-myc and P16gene proteins in human medulloblastoma
【摘要】 目的探讨EGFR、Nmyc、PCNA和P16基因在髓母细胞瘤发生中的作用,以及上述基因蛋白检测对髓母细胞瘤的诊断、病理分型和年龄的临床意义。方法用免疫组织化学LSAB法检测四种基因蛋白的产物在41例人脑髓母细胞瘤中的表达,分析与髓母细胞瘤病理分型和年龄间的关系。结果EGFR、Nmyc和PCNA蛋白在髓母细胞瘤中高度表达,P16蛋白有明显的缺失,分别为56.10%、60.98%、65.89%、58.54%。EGRF蛋白在两种主要病理类型中的表达差异有统计学意义(P<0.01),P16蛋白在两组不同年龄组间的差异有统计学意义(P<0.05)。结论上述四种基因在髓母细胞瘤的发生中可能起重要作用,EGFR和P16蛋白的检测对病理分型和预后判断有临床实际应用意义。
【Abstract】 Objective To investigate the significance of EGFR, N-myc, PCNA and P16 genes in the development, diagnosis and pathological classification of human medulloblastoma, we examined their expressions in tumor tissue. Methods The expressions of EGFR, N-myc, PCNA and P16 gene proteins were detected immunohistochemically in 41 patients with medulloblastoma. The relationship between pathological classifications and ages were determined. Results Expression of EGFR,N-myc and PCNA, deletion of P16 were 56.10%,60.98%,65.8% and 58.54% in human medulloblastoma respectively. It showed significant difference(P<0.01) between the two main pathological classifications in EGFR and between the two different age groups in p16(P<0.05). Conclusions The genes mentioned above may play an important role in the development of medulloblastoma, especially EGFR and P16. The detection of these proteins has a positive significance on malignancy determination of medulloblastoma.
