【摘要】 目的　探讨外源性前列腺素E1(PGE1)对血吸虫病家兔肝脏血小板衍生生长因子B(PDGF B)及其受体表达的影响。方法　14只血吸虫病纤维化家兔模型,其中7只家兔在感染后6 0d开始给予PGE1( 2 5 μg/kg·d-1)至12 0d。RT PCR、Westernblot和免疫组化分别检测肝组织PDGF BmRNA、PDGFR β蛋白和α平滑肌肌动蛋白(αSMA)表达。原位杂交观察内源性干扰素(IFN)γ水平。结果　纤维化形成过程中HSCs活化,PDGF BmRNA以及PDGFR β蛋白水平增高。外源性PGE1治疗后PDGFmRNA和受体β蛋白表达的增高趋势都受到明显抑制(与模型组比较分别为2 9 4 2±5 0 5和4 1 37±7 2 3,P <0 0 1;0 2 1±0 11和0 4 6±0 0 7,P <0 0 1)。αSMA表达强阳性视野从6 8下降到10个;而内源性IFN γ积分光密度从0 10 5±0 0 2 4升高到0 4 38±0 0 76 (P <0 0 1)。结论　PGE1可以降低血吸虫病家兔肝脏PDGF B基因及其受体表达水平,内源性IFN γ水平上调可能是这种作用的机制之一。更多还原

【Abstract】 Objective To investigate the effects of exogenous prostaglandin E 1 (PGE 1) on expression of platelet derived growth factor B (PDGF-B) mRNA and its receptor β protein in rabbit with schistosomiasis. Methods In this study, 14 rabbits were infected with cercaria of S. japonicum percutaneously. PGE 1 ( 2.5 μg/kg·d -1 ) was given intravenously to 7 rabbits from the 60th day to day 120. The expressions of PDGF-B mRNA, PDGFR β protein and α-SMA were detected by RT-PCR, Western blotting and immunohistochemistry, respectively. Endogenous IFN-γ was measured by in situ hybridization. Results Up-regulated expressions of PDGF-B mRNA, receptor β protein as well as α-SMA were observed in rabbits with Schistosome hepatic fibrosis. The increased expressions of PDGF mRNA and receptor β were suppressed in rabbits treated with exogenous PGE 1 (29.42±5.05 vs 41.37±7.23, P <0.01 and 0.21±0.11 vs 0.46±0.07, P <0.01). The expression of α-SMA significantly decreased, and mean O.D. of endogenous IFN-γ increased from 0.105±0.024 to 0.438±0.076 ( P <0.01). Conclusion Our results suggest that PGE 1 inhibits the expressions of PDGF-B gene and its receptor β effectively. Up-regulated endogenous IFN-γ may mediate in this progress.更多还原
